Fabp4-Cre-mediated Sirt6 deletion impairs adipose tissue function and metabolic homeostasis in mice
SIRT6 is a member of sirtuin family of deacetylases involved in diverse processes including genome stability, metabolic homeostasis and anti-inflammation. However, its function in the adipose tissue is not well understood. To examine the metabolic function of SIRT6 in the adipose tissue, we generated two mouse models that are deficient in Sirt6 using the Cre-lox approach. Two commonly used Cre lines that are driven by either the mouse Fabp4 or Adipoq gene promoter were chosen for this study. The Sirt6-knockout mice generated by the Fabp4-Cre line (Sirt6f/f:Fabp4-Cre) had a significant increase in both body weight and fat mass and exhibited glucose intolerance and insulin resistance as compared with the control wild-type mice. At the molecular levels, the Sirt6f/f :Fabp4-Cre-knockout mice had increased expression of inflammatory genes including F4/80, TNFα, IL-6 and MCP-1 in both white and brown adipose tissues. Moreover, the knockout mice showed decreased expression of the adiponectin gene in the white adipose tissue and UCP1 in the brown adipose tissue, respectively. In contrast, the Sirt6 knockout mice generated by the Adipoq-Cre line (Sirt6f/f :Adipoq-Cre) only had modest insulin resistance. In conclusion, our data suggest that the function of SIRT6 in the Fabp4-Cre-expressing cells in addition to mature adipocytes plays a critical role in body weight maintenance and metabolic homeostasis
Physical Activity, Screen Time, and Emotional Well-Being during the 2019 Novel Coronavirus Outbreak in China
We aimed to evaluate the effects of the COVID-19 lock down on lifestyle in China during the initial stage of the pandemic. A questionnaire was distributed to Chinese adults living in 31 provinces of China via the internet using a snowball sampling strategy. Information on 7-day physical activity recall, screen time, and emotional state were collected between January 24 and February 2, 2020. ANOVA, χ² test, and Spearman's correlation coefficients were used for statistical analysis. 12,107 participants aged 18-80 years were included. During the initial phase of the COVID-19 outbreak, nearly 60% of Chinese adults had inadequate physical activity (95% CI 56.6%-58.3%), which was more than twice the global prevalence (27.5%, 25.0%-32.2%). Their mean screen time was more than 4 hours per day while staying at home (261.3 ± 189.8 min per day), and the longest screen time was found in young adults (305.6 ± 217.5 min per day). We found a positive and significant correlation between provincial proportions of confirmed COVID-19 cases and negative affect scores (r = 0.501, p = 0.004). Individuals with vigorous physical activity appeared to have a better emotional state and less screen time than those with light physical activity. During this nationwide lockdown, more than half of Chinese adults temporarily adopted a sedentary lifestyle with insufficient physical activity, more screen time, and poor emotional state, which may carry considerable health risks. Predictive Value of Plasma MicroRNA-216a/b in the Diagnosis of Esophageal Squamous Cell Carcinoma
Esophageal squamous cell carcinoma (ESCC) is a common human malignancy with poor survival, which was usually diagnosed at an advanced stage. MicroRNAs (miRNAs), a class of single stranded noncoding RNAs with only 17-25 ribonucleotides, were demonstrated to play an important role in lots of cancers. In the recent years, increasing evidence revealed that circulating miRNAs exhibited great potential in the diagnosis of various types of cancers. The present study was designed to evaluate the diagnostic value of plasma miRNA-216a/b for ESCC. Our results showed that the expression level of plasma miRNA-216a/b was significantly lower in ESCC patients compared with that of healthy controls. The receiver operating characteristic (ROC) curve analysis yielded an area under the ROC curve (AUC) value of 0.877 [95% CI (confidence interval): 0.818-0.922] for miRNA-216a and 0.756 (95% CI: 0.685-0.819) for miRNA-216b. Clinical data indicated that plasma miRNA-216a/b were inversely correlated with lymph node metastasis and TNM stage. Additionally, the plasma miRNA-216b expression level was significantly upregulated in postoperative samples compared to preoperative samples. Our study, for the first time, demonstrated that plasma miRNA-216a/b might serve as potential biomarkers for the diagnosis of ESCC and dysregulation of miRNA-216a/b might be involved in the progression of ESCC
TEPA modification Cu-BTC@SiO2 preparation of composite aerogel and its CO2 capture characteristics
Under the dual strategic background of “carbon peaking and carbon neutrality”, CO2 capture has become an important task at present. Solid adsorbent adsorption is widely used in CO2 capture process, among which SiO2 aerogel has the advantages of low cost, flexible synthesis method, high separation efficiency, easy surface modification, etc. However, SiO2 aerogel materials also have some defects, such as low CO2/N2 adsorption selectivity and CO2 adsorption capacity to be further improved. To address the above issues, this article has prepared a Cu-BTC@SiO2 Composite aerogel CO2 adsorption material. Firstly, the surface chemistry and pore structure were systematically characterized by scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), and nitrogen adsorption and desorption tests. Then, the CO2 adsorption capacity, selective adsorption, and cyclic adsorption were studied through carbon dioxide adsorption testing. Finally, a combination of theoretical and experimental research was used to study the CO2 adsorption kinetics of the adsorbent. The results show that the SiO2 aerogel compounded with Cu BTC has a high specific surface area of 726.431 m2/g, a specific surface area of 570.781 m2/g, and a high microporous volume of 0.184 cm3/g. After loading tetraethylenepentamine(TEPA), the adsorption capacity of CO2 is up to 2.95 mmol/g, and the selective adsorption is 40.8, after 10 cycles of CO2 adsorption, the adsorption capacity decreased slightly. Therefore, TEPA-modified Cu-BTC@SiO2 composite aerogels can significantly improve the CO2 adsorption performance of SiO2 aerogels. The metal organic framework material Cu BTC with rich micropore structure is compounded with SiO2 aerogel, and is prepared by the sol gel method Cu-BTC@SiO2 Composite aerogel to make the composite have hierarchical micro/mesoporous structure and enhance the physical adsorption of CO2 by enhancing the intermolecular force (van der Waals force); The material is impregnated with TEPA, and the chemical adsorption of CO2 is enhanced by acid-base interaction between organic amine and acid gas
Factor Structure of the Chinese Version of the Parent Adult-Child Relationship Questionnaire
The Parent Adult-Child Relationship Questionnaire (PACQ) included two identical versions of the 13-item scale, which were administered to each subject, one which referred to “relationship with mother” and the other to “relationship with father.” The PACQ, originally in English, is a self-report measure of the filial relationship. The present study aimed to develop a Chinese version of the PACQ and use it to explore Chinese parent adult-child relationships. A total of 454 Chinese adult-children completed the Chinese version of the PACQ. The structure of the questionnaire was analyzed using exploratory factor analysis (EFA) and confirmatory factor analysis (CFA). We found that the Cronbach's α was 0.66–0.88 for fathers and 0.76–0.91 for mothers, which demonstrates high internal consistency reliabilities of the Chinese version of the PACQ. The Chinese version of the PACQ for father had similar constructs similar to with those of the original English version. However, a new factor for mothers, “attachment,” was derived from the original English version. The results suggested that the Chinese version of PACQ is a valid and reliable measure of relationship quality between Chinese adult-children and their parents

Integrative analysis of key candidate genes and signaling pathways in ovarian cancer by bioinformatics
Abstract Background Ovarian cancer is one of the most common gynecological tumors, and among gynecological tumors, its incidence and mortality rates are fairly high. However, the pathogenesis of ovarian cancer is not clear. The present study aimed to investigate the differentially expressed genes and signaling pathways associated with ovarian cancer by bioinformatics analysis. Methods The data from three mRNA expression profiling microarrays (GSE14407, GSE29450, and GSE54388) were obtained from the Gene Expression Omnibus (GEO) database. Differentially expressed genes between ovarian cancer tissues and normal tissues were identified using R software. The overlapping genes from the three GEO datasets were identified, and profound analysis was performed. The overlapping genes were used for pathway and Gene Ontology (GO) functional enrichment analysis using the Metascape online tool. Protein–protein interactions were analyzed with the Search Tool for the Retrieval of Interacting Genes/Proteins (STRING). Subnetwork models were selected using the plugin molecular complex detection (MCODE) application in Cytoscape. Kaplan–Meier curves were used to analyze the univariate survival outcomes of the hub genes. The Human Protein Atlas (HPA) database and Gene Expression Profiling Interactive Analysis (GEPIA) were used to validate hub genes. Results In total, 708 overlapping genes were identified through analyses of the three microarray datasets (GSE14407, GSE29450, and GSE54388). These genes mainly participated in mitotic sister chromatid segregation, regulation of chromosome segregation and regulation of the cell cycle process. High CCNA2 expression was associated with poor overall survival (OS) and tumor stage. The expression of CDK1, CDC20, CCNB1, BUB1B, CCNA2, KIF11, CDCA8, KIF2C, NDC80 and TOP2A was increased in ovarian cancer tissues compared with normal tissues according to the Oncomine database. Higher expression levels of these seven candidate genes in ovarian cancer tissues compared with normal tissues were observed by GEPIA. The protein expression levels of CCNA2, CCNB1, CDC20, CDCA8, CDK1, KIF11 and TOP2A were high in ovarian cancer tissues, which was further confirmed via the HPA database. Conclusion Taken together, our study provided evidence concerning the altered expression of genes in ovarian cancer tissues compared with normal tissues. In vivo and in vitro experiments are required to verify the results of the present study
