    Synthetic molecules and functionalized nanoparticles targeting the LPS-TLR4 signaling: A new generation of immunotherapeutics

    Toll-like receptor 4 (TLR4), the receptor of bacterial endotoxins in mammalians, plays a pivotal role in the induction of innate immunity and inflammation. TLR4 activation by bacterial lipopolysaccharide (LPS) is achieved by the coordinate and sequential action of three other proteins, the lipopolysaccharide binding protein (LBP), the cluster differentiation antigen CD14, and the myeloid differentiation protein (MD-2) receptors, that bind LPS and present it in a monomeric form to TLR4 by forming the activated [TLR4·MD-2·LPS]2 complex. Small molecules and nanoparticles active in modulating the TLR4 signal by targeting directly the MD-2·TLR4 complex or by interfering in other points of the TLR4 signaling are presented in this paper. These compounds have great pharmacological interest as vaccine adjuvants, immunotherapeutics, anti-sepsis, and anti-inflammatory agents

    Constraining Modified Gravity with Euclid

    Future proposed satellite missions as Euclid can offer the opportunity to test general relativity on cosmic scales through mapping of the galaxy weak lensing signal. In this paper we forecast the ability of these experiments to constrain modified gravity scenarios as those predicted by scalar-tensor and f(R)f(R) theories. We found that Euclid will improve constraints expected from the PLANCK satellite on these modified gravity models by two orders of magnitude. We discuss parameter degeneracies and the possible biases introduced by modified gravity

    AAA: Fair Evaluation for Abuse Detection Systems Wanted

    User-generated web content is rife with abusive language that can harm others and discourage participation. Thus, a primary research aim is to develop abuse detection systems that can be used to alert and support human moderators of online communities. Such systems are notoriously hard to develop and evaluate. Even when they appear to achieve satisfactory performance on current evaluation metrics, they may fail in practice on new data. This is partly because datasets commonly used in this field suffer from selection bias, and consequently, existing supervised models overrely on cue words such as group identifiers (e.g., gay and black) which are not inherently abusive. Although there are attempts to mitigate this bias, current evaluation metrics do not adequately quantify their progress. In this work, we introduce Adversarial Attacks against Abuse (AAA), a new evaluation strategy and associated metric that better captures a model’s performance on certain classes of hard-to-classify microposts, and for example penalises systems which are biased on low-level lexical features. It does so by adversarially modifying the model developer’s training and test data to generate plausible test samples dynamically. We make AAA available as an easy-to-use tool, and show its effectiveness in error analysis by comparing the AAA performance of several state-of-the-art models on multiple datasets. This work will inform the development of detection systems and contribute to the fight against abusive language online

    Exploring the soft shades of sustainability. Evidence from Italian companies.

    Purpose – Sustainability is increasingly at the forefront of the public debate in Europe and the world. However, despite this increased interest, research seems to have partially ignored the importance of its social dimension and the issues related to social equity, people care, protection, and personal development at all stages of society and, consequently, of business. Accordingly, this paper aims at investigating the “soft” dimensions of sustainability, integrating its mainstream “technical storyline” with a “human/social storyline”. Design/methodology/approach – A taxonomy of the main key drivers of the soft dimension of sustainability is proposed and tested on a sample of Italian companies. Through interviews with their managers, actions and needs in terms of sustainability soft drivers are identified. Findings – The achieved results demonstrated that the case companies differently integrated the soft dimensions of sustainability within their companies. All the sample companies are aware of the role of social sustainability. According to the proposed taxonomy, the systemic drivers of soft sustainability are the main shared ones. Originality/value – The paper provides new insights into the essence of the organizational soft dimensions and their centrality in the overall achievement of sustainability for companies. It also offers managerial insights into how to effectively manage these dimensions and policy implications about the need for clearer consideration

    Perfusion index variations in clinically and hemodynamically stable preterm newborns in the first week of life

    <p>Abstract</p> <p>Background</p> <p>The perfusion index, derived from the pulse oximeter signal, seems to be an accurate predictor for high illness severity in newborns. The aim of this study was to determine the perfusion index values of clinically and hemodynamically stable preterm newborns in the first week of life.</p> <p>Methods</p> <p>Perfusion index recordings were performed on the first, third and seventh day of life on 30 preterm newborns. Their state of health was assessed according to clinical and behaviour evaluation and to the Score for Neonatal Acute Physiology.</p> <p>Results</p> <p>The median(interquartile range) perfusion index values were 0.9(0.6) on the first, 1.2(1.0) on the third, and 1.3(0.9) on the seventh day, with a significant increase between the first and the third day.</p> <p>Conclusions</p> <p>Perfusion index proved to be an easily applicable, non-invasive method for monitoring early postnatal changes in peripheral perfusion. Its trend during the first week of life suggests that its clinical application should take age into account. Further studies are needed to obtain reference perfusion index values from a larger sample of preterm newborns, to identify specific gestational age-related cut-off values for illness and to test the role of perfusion index in monitoring critically ill neonates.</p

    Healthspan Enhancement by Olive Polyphenols in C. elegans Wild Type and Parkinson’s Models

    Parkinson’s disease (PD) is the second most prevalent late-age onset neurodegenerative disorder, affecting 1% of the population after the age of about 60 years old and 4% of those over 80 years old, causing motor impairments and cognitive dysfunction. Increasing evidence indicates that Mediterranean diet (MD) exerts beneficial effects in maintaining health, especially during ageing and by the prevention of neurodegenerative disorders. In this regard, olive oil and its biophenolic constituents like hydroxytyrosol (HT) have received growing attention in the past years. Thus, in the current study we test the health-promoting effects of two hydroxytyrosol preparations, pure HT and HidroxÂź (HD), which is hydroxytyrosol in its “natural” environment, in the established invertebrate model organism Caenorhabditis elegans. HD exposure led to much stronger beneficial locomotion effects in wild type worms compared to HT in the same concentration. Consistent to this finding, in OW13 worms, a PD-model characterized by α-synuclein expression in muscles, HD exhibited a significant higher effect on α-synuclein accumulation and swim performance than HT, an effect partly confirmed also in swim assays with the UA44 strain, which features α-synuclein expression in DA-neurons. Interestingly, beneficial effects of HD and HT treatment with similar strength were detected in the lifespan and autofluorescence of wild-type nematodes, in the neuronal health of UA44 worms as well as in the locomotion of rotenone-induced PD-model. Thus, the hypothesis that HD features higher healthspan-promoting abilities than HT was at least partly confirmed. Our study demonstrates that HD polyphenolic extract treatment has the potential to partly prevent or even treat ageing-related neurodegenerative diseases and ageing itself. Future investigations including mammalian models and human clinical trials are needed to uncover the full potential of these olive compounds.Peer Reviewe
