    ATP depletion induces translocation of STIM1 to puncta and formation of STIM1–ORAI1 clusters: translocation and re-translocation of STIM1 does not require ATP

    Depletion of the endoplasmic reticulum (ER) calcium store triggers translocation of stromal interacting molecule one (STIM1) to the sub-plasmalemmal region and formation of puncta—structures in which STIM1 interacts and activates calcium channels. ATP depletion induced the formation of STIM1 puncta in PANC1, RAMA37, and HeLa cells. The sequence of events triggered by inhibition of ATP production included a rapid decline of ATP, depletion of phosphatidylinositol 4,5-bisphosphate (PI(4,5)P2) and a slow calcium leak from the ER followed by formation of STIM1 puncta. STIM1 puncta induced by ATP depletion were co-localized with clusters of ORAI1 channels. STIM1–ORAI1 clusters that developed as a result of ATP depletion were very poor mediators of Ca2+ influx. Re-translocation of STIM1 from puncta back to the ER was observed during total ATP depletion. We can therefore conclude that STIM1 translocation and re-translocation as well as formation of STIM1–ORAI1 clusters occur in an ATP-independent fashion and under conditions of PI(4,5)P2 depletion

    Microsatellite mapping of a gene affecting horn development in Bos taurus

    The presence or absence of horns in Bos taurus is thought to be under the genetic control of the autosomal polled locus, characterized by two alleles: P dominant over p, and causing the polled or hornless phenotype. We have demonstrated genetic linkage between the polled locus and two microsatellite markers, GMPOLL-1 and GMPOLL-2, and have assigned the corresponding linkage group to bovine chromosome 1. This confirms the existence of the postulated polled locus and the hypothesized inheritance pattern. It will allow marker assisted selection for the polledness trait in breeding programs and is a first step towards positional cloning and molecular study of a gene that has been subjected to both natural and artificial selection

    Brightest Cluster Galaxy Formation in the z = 4.3 Protocluster SPT 2349-56: Discovery of a Radio-loud Active Galactic Nucleus

    We have observed the z = 4.3 protocluster SPT2349−56 with the Australia Telescope Compact Array (ATCA) with the aim of detecting radio-loud active galactic nuclei (AGNs) among the ∼30 submillimeter (submm) galaxies (SMGs) identified in the structure. We detect the central complex of submm sources at 2.2 GHz with a luminosity of L _2.2 = (4.42 ± 0.56) × 10 ^25 W Hz ^−1 . MeerKAT and the Australian Square Kilometre Array Pathfinder also detect the source at 816 MHz and 888 MHz, respectively, constraining the radio spectral index to α = −1.45 ± 0.16, implying L _1.4,rest = (2.2 ± 0.2) × 10 ^26 W Hz ^−1 . The radio observations do not have sufficient spatial resolution to uniquely identify one of the three Atacama Large Millimeter/submillimeter Array (ALMA) galaxies as the AGN, however the ALMA source properties themselves suggest a likely host. This radio luminosity is ∼100× higher than expected from star formation, assuming the usual far-infrared–radio correlation, indicating an AGN driven by a forming brightest cluster galaxy. None of the SMGs in SPT2349−56 show signs of AGNs in any other diagnostics available to us, highlighting the radio continuum as a powerful probe of obscured AGNs. We compare these results to field samples of radio sources and SMGs, along with the 22 gravitationally lensed SPT-SMGs also observed in the ATCA program, as well as powerful radio galaxies at high redshifts. The (3.3 ± 0.7) × 10 ^38 W of power from the radio-loud AGN sustained over 100 Myr is comparable to the binding energy of the gas mass of the central halo, and similar to the instantaneous energy injection from supernova feedback from the SMGs in the core region. The SPT2349−56 radio-loud AGNs may be providing strong feedback on a nascent intracluster medium

    Labor force participation and health-related quality of life in HIV-positive men who have sex with men: The multicenter AIDS cohort study

    Too many people with HIV have left the job market permanently and those with reduced work capacity have been unable to keep their jobs. There is a need to examine the health effects of labor force participation in people with HIV. This study presents longitudinal data from 1,415 HIV-positive men who have sex with men taking part in the Multicenter AIDS Cohort Study. Generalized Estimating Equations show that employment is associated with better physical and mental health quality of life and suggests that there may be an adaptation process to the experience of unemployment. Post hoc analyses also suggest that people who are more physically vulnerable may undergo steeper health declines due to job loss than those who are generally healthier. However, this may also be the result of a selection effect whereby poor physical health contributes to unemployment. Policies that promote labor force participation may not only increase employment rates but also improve the health of people living with HIV. © Springer Science+Business Media, LLC 2012
