    Subjective response to alcohol and ADH polymorphisms in a select sample of young adult male East Indians and Africans in Trinidad and Tobago

    OBJECTIVE: Level of response to alcohol has been associated with risk of alcohol dependence in a number of ethnic groups. In the present study, subjective and objective responses to alcohol were evaluated in Indo-Trinidadians (Indo-T) and Afro-Trinidadians (Afro-T). Associations of alcohol dehydrogenase polymorphisms with response to alcohol, using the Subjective High Assessment Scale (SHAS), and breath alcohol concentrations (BrAC) were tested. METHOD: Regular male drinkers without alcohol dependence (n = 112) ages 18-25 years participated in alcohol challenge sessions consisting of placebo and two doses of alcohol (target BrAC: 0 g/dl for placebo, .04 g/dl low dose, and .08 g/dl high dose) and genotyped for variants in ADH1B*3 and ADH1C*2. RESULTS: Indo-T had significantly higher BrAC, pulse rates, and cortisol levels when compared with Afro-T but did not have significantly higher SHAS values. Higher responses on the SHAS items muddle/confused and nauseated were significantly associated with the presence of at least one ADH1B*3 allele following the high dose of alcohol in Afro-T. Indo-T with at least one ADH1C*2 allele displayed significantly different Drug × Time interactions for the SHAS item effects of alcohol at the low dose and for the SHAS items clumsy, muddle/confused, effects of alcohol, floating, drunk, and total at the high dose from Indo-T with two ADH1C*1 alleles. CONCLUSIONS: This is the first study that has investigated individual sensitivity to alcohol in a Caribbean population and in people of East Indian descent. Indo-T with at least one ADH1C*2 allele may be at higher risk for heavy drinking by feeling less of the effects of alcohol, including nausea. In Afro-T, having at least one ADH1B*3 allele appears to exert a protective effect by enhancing the unpleasant effects of alcohol, such as nausea and confusion

    Optimizing Glycerin-to-water Ratios for Preserving Eucalyptus Foliage

    Fresh eucalyptus stems can be preserved via systemic absorption of glycerin, but the optimal rate of glycerin and its effect on foliage color and stem pliability during holding have not been well-described. We harvested stems of Eucalyptus pulverulenta ‘Baby Blue’ and treated them in solutions of 0% (control), 25%, 50%, and 75% glycerin:reverse-osmosis water for 2 weeks. Leaf greenness was measured nine times during this period; the change in stem weights and solution uptake were measured as well. Stem pliability was evaluated after 2 weeks in solution as well as 1 week after holding the stems dry. The 25% and 50% glycerin treatments increased stem weight, whereas the control and 75% treatments decreased stem weight. The glycerin treatments absorbed less than 50 mL solution per three stems during 2 weeks, whereas those in the control absorbed >270 mL water. The color of foliage was similar during the first 5 days in treatment solutions, but stems in 25% and 50% glycerin treatments were less green than those in the control and 75% treatments by day 6, and this trend continued. No difference occurred in stem pliability among treatments after 2 weeks in glycerin solutions; however, after 1 week of holding, the top 9 cm of stems in glycerin treatments were more pliable than those in the control. Based on these results, we recommend a rate of 25% glycerin to preserve eucalyptus foliage for use in botanical artistry

    FAMILIAL DYSALBUMINAEMIC HYPERTHYROXINEMIA INTERFERES WITH CURRENT FREE THYROID HORMONE IMMUNOASSAY METHODS

    Familial dysalbuminaemic hyperthyroxinemia (FDH), most commonly due to an Arginine to Histidine mutation at residue 218 (R218H) in the albumin gene, causes artefactual elevation of free thyroid hormones in euthyroid individuals. We have evaluated the susceptibility of most current free thyroid hormone immunoassay methods used in the UK, Europe and Far East to interference by R218H FDH. Methods: Different, one- and two-step immunoassay methods were tested, measuring Free T4 (FT4) and Free T3 (FT3) in 37 individuals with genetically-proven R218H FDH. Results: With the exception of Ortho VITROS, FT4 measurements were raised in all assays, with greatest to lowest susceptibility to interference being Beckman ACCESS > Roche ELECSYS > FUJIREBIO Lumipulse > Siemens CENTAUR > Abbott ARCHITECT > Perkin-Elmer DELFIA. Five different assays recorded high FT3 levels, with the Siemens CENTAUR method measuring high FT3 values in up to 30% of cases. However, depending on the assay method, FT4 measurements were unexpectedly normal in some, genetically-confirmed, affected relatives of index FDH cases. Conclusions: All FT4 immunoassays evaluated are prone to interference by R218H FDH, with their varying susceptibility not being related to assay architecture but likely due to differing assay conditions or buffer composition. Added susceptibility of many FT3 assays to measurement interference, resulting in high FT4 and FT3 with non-suppressed TSH levels, raises the possibility of R218H FDH being misdiagnosed as Resistance to Thyroid Hormone beta or TSH-secreting pituitary tumour, potentially leading to unnecessary investigation and inappropriate treatment.Research is supported by funding from the Wellcome Trust (210755/Z/18/Z to KC) and NIHR Cambridge Biomedical Research Centre (CM, MG, KC)

    Ecologia de ervas-de-passarinho Neotropicais: um importante componente do dossel de ecossistemas brasileiros

    Mistletoes have been studied in temperate countries regularly because they can be pests of cultivated plants and forest plantations. In comparison with temperate habitats, little is known about the ecology of mistletoes in the Neotropics. More emphasis should be given to neotropical mistletoes because they could be important elements of plant communities, acting as key resources for pollinators, seed dispersers and herbivores. Using a combination of findings from early mistletoe studies and empirical evidence from several recent case studies, we report emerging patterns of mistletoe interactions with associated organisms and propose aims for further studies. There has been increasing interest in working with mistletoes in Brazil. Although there are some data on the ecology of mistletoe seed dispersal, reproductive biology studies are scarce and represent a wide range of fieldwork to be explored. Knowledge of the basic biology of mistletoes will be very important for modeling their spatial distribution using metapopulation or epidemiological approaches. In this review, we summarize the studies conducted in the neotropical region in order to provide a framework for current research and new ideas for future investigations of mistletoes, especially in Brazil.Ervas-de-passarinho têm sim sido regularmente estudadas em países temperados por afetar negativamente espécies cultivadas e florestas manejadas.     Managing health care in the digital world:A comparative analysis

    Recently, most reforms affecting healthcare systems have focused on improving the quality of care and containing costs. This has led many scholars to advocate the adoption of Health Information systems, especially electronic medical records, by highlighting their potential benefits. This study is based on a comparative analysis using a multiple method approach to examine the implementation of the same electronic medical record system at two different hospitals. Its findings offer insights into the processes of the adoption of innovation and its implementation in a healthcare context. The need to innovate, the decision to innovate, the implementation process and consequently, the results produced are quite distinctive at each study site. This comparative case study reveals that what appears to be the same can be quite different: this can be due to several conditions at the organization, the organization’s characteristics, and the process of implementation adopted. We need to understand these elements in order to be able to plan and manage such programs in the future

    The Helicobacter pylori Genome Project : insights into H. pylori population structure from analysis of a worldwide collection of complete genomes

    Helicobacter pylori, a dominant member of the gastric microbiota, shares co-evolutionary history with humans. This has led to the development of genetically distinct H. pylori subpopulations associated with the geographic origin of the host and with differential gastric disease risk. Here, we provide insights into H. pylori population structure as a part of the Helicobacter pylori Genome Project (HpGP), a multi-disciplinary initiative aimed at elucidating H. pylori pathogenesis and identifying new therapeutic targets. We collected 1011 well-characterized clinical strains from 50 countries and generated high-quality genome sequences. We analysed core genome diversity and population structure of the HpGP dataset and 255 worldwide reference genomes to outline the ancestral contribution to Eurasian, African, and American populations. We found evidence of substantial contribution of population hpNorthAsia and subpopulation hspUral in Northern European H. pylori. The genomes of H. pylori isolated from northern and southern Indigenous Americans differed in that bacteria isolated in northern Indigenous communities were more similar to North Asian H. pylori while the southern had higher relatedness to hpEastAsia. Notably, we also found a highly clonal yet geographically dispersed North American subpopulation, which is negative for the cag pathogenicity island, and present in 7% of sequenced US genomes. We expect the HpGP dataset and the corresponding strains to become a major asset for H. pylori genomics
