IDENTIFIKASI TELUR CACING ASCARIS LUMBRICOIDES PADA KEMANGI (OCIMUM BASILICUM L) YANG DIJUAL DI PASAR

Ascaris lumbricoides is a roundworm which is a class of intestinal nematodes that are commonly found in tropical and subtropical areas where the condition of the area shows a low level of environmental hygiene. The prevalence of worm egg infection is generally transmitted through food, drinks and vegetables such as basil which is consumed raw as vegetables that are not clean in washing. This study aims to identify the eggs of Ascaris Lumbricoides worm in basil vegetables with samples obtained from the market. Experimental research method with qualitative laboratory examination with Flotation method to identify worm eggs. The population of all Basil vegetables are sold from vegetable traders in the market. Sampling is all basil vegetables from the market. Primary data generated from basil samples were observed. From the results of research conducted on basil vegetables showed negative results containing worm eggs, so basil is safe for consumption.  Keywords: Ascaris lumbricoides, Basil, Worm egg

Understanding the Health-related Quality of Life of People Living with HIV Based on Sexual Orientation

Human immunodeficiency virus (HIV) infection has detrimental impacts on the lives of different population groups living with HIV, including men who have sex with men (MSM). Using the World Health Organization Quality of Life questionnaire, this study aimed to assess the health-related quality of life of men living with HIV with different sexual orientations and to determine the dominant influential factors. This cross-sectional study involved 206 men living with HIV. They were recruited from the Sriwijaya Plus Foundation and a medical facility that provided antiretroviral therapy. The data were analyzed using Chi-square and binomial logistic regression. The analysis showed that the percentage of MSM patients was greater than that of non-MSM patients, accounting for 68.9% of the total population. The multivariate logistic regression revealed that the most dominant influential factor was depression status (PR = 5.417; 95% CI = 2.473–11.876), with the majority of the depressed patients being 5.417 times more at risk of a lower quality of life compared to others. These findings suggest that depression can lead to a low quality of life among HIV patients

<i>In Silico</i> and Validation Approaches for Optimum Conditions of <i>Rattus norvegicus</i> Target Gene qPCR Primers

The qPCR method requires an oligonucleotide pair to prime the amplification process. With the variety of qPCR reagent and primer options available, in silico and laboratory experimental validation approach was needed to validate the most suitable primer for prior use. This article aims to provide in silico analysis of actin alpha-2 smooth muscle (Acta2), fibroblast activation protein (Fap), hypoxanthine phosphoribosyltransferase-1 (Hprt1), platelet-derived growth factor subunit B (Pdgfb), phosphoinositide-3-kinase regulatory subunit-1 (Pik3r1), and vascular cell adhesion molecule-1 (Vcam1) qPCR primer with qPCR and electrophoresis validation. The procedure used in this approach was in silico analysis of primer from published articles and newly designed primer. The analysis was done with Primer-BLAST for gene specificity, Primer-Dimer, OligoCalc for hairpin formation, BLAST Nucleotide for identical sequence screening, and Clustal Omega for product length validation. Experimental validation was done using qPCR for optimal annealing temperature, priming ability, and amplificon specificity, and electrophoresis for product length validation. This assessment resulted in in silico and laboratory experimental validation of Acta2, Fap, Hprt1, Pdgfb, Pik3r1, and Vcam1 primer pairs producing suitable amplicon for qPCR using Rattus norvegicus cDNA with SYBR annealing temperature range of 60-65°C with three mM MgCl2. The primer pair can be used for further qPCR analysis under similar conditions and the procedure stated can be used as starting point for qPCR Primer preparation

Dalam pelaksanaan pembangunan ekonomi, perekonomian memiliki peran dan kepentingan penting sebagai faktor pendukung keberhasilan pembangunan nasional, dan menjaga kerjasama dalam upaya memperluas skala pembangunan ekonomi adalah aset yang paling penting. Dengan latar belakang masalah di atas, beberapa pertanyaan dapat dipertimbangkan sebagai berikut: Ciri-ciri Perjanjian Kerjasama Bagaimana program gerakan sekolah menulis buku nasional antara PT Nyala Masa depan Indonesia dan SMP Negeri 1 Denpasar, dan bagaimana jika ada Perjanjian Kerjasama antara PT Nyala Masadepan Indonesia dan SMP Negeri 1 Denpasar Program gerakan sekolah telah didirikan untuk menulis buku secara nasional. Metode yang digunakan adalah penelusuran hukum normatif dengan menggunakan sumber bahan hukum primer dan sekunder. Hasil survei ini menyatakan: Karakteristik perjanjian kerjasama antara PTs. Indonesian Future Light dan SMP Negeri 1 Denpasar menggunakan kesepakatan baku. Model kontrak berupa kontrak tertulis. Kegagalan perjanjian kerjasama antara PT. Nyala Masa depan Indonesia dan SMP Negeri 1 Denpasar dilakukan melalui proses dan proses non prosedural.
