An enzyme-based screening system for the rapid assessment of protein N-glycosylation efficiency in yeast
N-Glycosylation efficiency is a key parameter when studying components of the protein N-glycosylation pathway, but was recently also recognized as an important factor in the production of glycosylated proteins. We have developed a novel assay to quantify N-glycosylation efficiency of proteins. This assay is based on the secreted activity of yeast acid phosphatase, the proper folding and hence secretion of which is strongly dependent on its N-glycosylation status. The results show that the reporter yields a quantitative measure for protein N-glycosylation in yeast, which is in good agreement with classically used assay based on protein migration patterns on SDS-PAGE. Expression of the DNA mismatch repair proteins hMLH1 and hPMS2 in normal human tissues.
hMLH1 and hPMS2 are part of the DNA mismatch repair complex. Mutations in these genes have been linked to hereditary non-polyposis colon cancer; they also occur in a variety of sporadic cancers. Western blot analysis and immunohistochemistry demonstrated that hMLH1 and hPMS2 are widely expressed nuclear proteins with a distribution pattern very similar to that previously described for hMSH2. These observations showing similar localization of hMLH1 and hPMS2 with hMSH2 are consistent with the biochemical function of these proteins in DNA mismatch repair
Local Real-Space View of the Achiral 1-TiSe 2 2 2 Charge Density Wave
The transition metal dichalcogenide 1-TiSe is a quasi-two-dimensional
layered material undergoing a commensurate 2 2 2 charge
density wave (CDW) transition with a weak periodic lattice distortion (PLD)
below 200 K. Scanning tunneling microscopy (STM) combined with
intentionally introduced interstitial Ti atoms allows to go beyond the usual
spatial resolution of STM and to intimately probe the three-dimensional
character of the PLD. Furthermore, the inversion-symmetric, achiral nature of
the CDW in the -direction is revealed, contradicting the claimed existence
of helical CDW stacking and associated chiral order. This study paves the way
to a simultaneous real-space probing of both charge and structural
reconstructions in CDW compounds.Comment: 5 pages, 4 figure
STM microscopy of the CDW in 1T-TiSe2 in the presence of single atom defects
We present a detailed low temperature scanning tunneling microscopy study of
the commensurate charge density wave (CDW) in 1-TiSe in the presence of
single atom defects. We find no significant modification of the CDW lattice in
single crystals with native defects concentrations where some bulk probes
already measure substantial reductions in the CDW phase transition signature.
Systematic analysis of STM micrographs combined with density functional theory
modelling of atomic defect patterns indicate that the observed CDW modulation
lies in the Se surface layer. The defect patterns clearly show there are no
2-polytype inclusions in the CDW phase, as previously found at room
temperature [Titov A.N. et al, Phys. Sol. State 53, 1073 (2011). They further
provide an alternative explanation for the chiral Friedel oscillations recently
reported in this compound [J. Ishioka et al., Phys. Rev. B 84, 245125, (2011)].Comment: 5 pages, 4 figure
Electronic Structure of the YH3 Phase from Angle-Resolved Photoemission Spectroscopy
Yttrium can be loaded with hydrogen up to high concentrations causing
dramatic structural and electronic changes of the host lattice. We report on
angle-resolved photoemission experiments of the Y trihydride phase. Most
importantly, we find the absence of metal d-bands at the Fermi level and a set
of flat, H-induced bands located at much higher binding energy than predicted,
indicating an increased electron affinity at H sites
A European spine registry
Abstract.: New joint replacement registries are being widely implemented across national and international organizations. The need for a continuous long-term post-market surveillance of implants has been recognized, and has become increasingly important to ensure the quality of prosthetic components. Registry data with large case numbers represent an acceptable alternative to controlled randomized clinical trials, which are often difficult to conduct in orthopedic surgery. The variety of implants and procedures in spinal surgery not only induces the same need for long-term monitoring of post-surgical product performance as in the joint replacement subspecialties, but also renders essential the establishment of a comprehensive spine registry for all major pathologies and interventions. In cooperation with the M.E. Müller Institute for Evaluative Research in Orthopedic Surgery (MEM-CED) at the University of Berne, Switzerland, the Spine Society of Europe (SSE) has launched Spine Tango: the first modular and multilevel European online registry for spinal surgery. Within Spine Tango, the major challenge in registry design and structure is the definition of and agreement upon a core set of questions as a common European dataset. Additional questions for national or individual interest can also be dynamically added to the core dataset. An automated implant tracking system has also been setup, which allows highly precise product documentation without additional work for clinical staff member
Heterogeneity in testing practices for infections during pregnancy: national survey across Switzerland.
QUESTION: Detection and treatment of infections during pregnancy are important for both maternal and child health. The objective of this study was to describe testing practices and adherence to current national guidelines in Switzerland.
METHODS: We invited all registered practicing obstetricians and gynaecologists in Switzerland to complete an anonymous web-based questionnaire about strategies for testing for 14 infections during pregnancy. We conducted a descriptive analysis according to demographic characteristics.
RESULTS: Of 1138 invited clinicians, 537 (47.2%) responded and 520 (45.6%) were eligible as they are currently caring for pregnant women. Nearly all eligible respondents tested all pregnant women for group B streptococcus (98.0%), hepatitis B virus (HBV) (96.5%) and human immunodeficiency virus (HIV) (94.7%), in accordance with national guidelines. Although testing for toxoplasmosis is not recommended, 24.1% of respondents tested all women and 32.9% tested at the request of the patient. Hospital doctors were more likely not to test for toxoplasmosis than doctors working in private practice (odds ratio [OR] 2.52, 95% confidence interval [CI] 1.04-6.13, p = 0.04). Only 80.4% of respondents tested all women for syphilis. There were regional differences in testing for some infections. The proportion of clinicians testing all women for HIV, HBV and syphilis was lower in Eastern Switzerland and the Zurich region (69.4% and 61.2%, respectively) than in other regions (range 77.1-88.1%, p <0.001). Most respondents (74.5%) said they would appreciate national guidelines about testing for infections during pregnancy.
CONCLUSIONS: Testing practices for infections in pregnant women vary widely in Switzerland. More extensive national guidelines could improve consistency of testing practices
Benchmarking with Spine Tango: potentials and pitfalls
The newly released online statistics function of Spine Tango allows comparison of own data against the aggregated results of the data pool that all other participants generate. This comparison can be considered a very simple way of benchmarking, which means that the quality of what one organization does is compared with other similar organizations. The goal is to make changes towards better practice if benchmarking shows inferior results compared with the pool. There are, however, pitfalls in this simplified way of comparing data that can result in confounding. This means that important influential factors can make results appear better or worse than they are in reality and these factors can only be identified and neutralized in a multiple regression analysis performed by a statistical expert. Comparing input variables, confounding is less of a problem than comparing outcome variables. Therefore, the potentials and limitations of automated online comparisons need to be considered when interpreting the results of the benchmarking procedur
