    Optical spectroscopy of radio galaxies in the 7C Redshift Survey

    We present optical spectroscopy of all 49 radio galaxies in the 7C-I and 7C-II regions of the 7C Redshift Survey (7CRS). The low-frequency (151 MHz) selected 7CRS sample contains all sources with flux-densities S_151 > 0.5 Jy in three regions of the sky; 7C-I and 7C-II were chosen to overlap with the 5C6 and 5C7 surveys respectively, and cover a total sky area of 0.013 sr. The sample has been completely identified and spectroscopy of the quasars and broad-lined radio galaxies has been presented in Willott et al. (1998). Only seven of the radio galaxies do not have redshift determinations from the spectroscopy, giving a redshift completeness for the sample of >90%. The median redshift of the 7CRS is 1.1. We present a composite 0.2<z<0.8 7CRS radio galaxy spectrum and investigate the strengths of the 4000 Angstrom breaks in these radio galaxies. We find an anti-correlation between the 4000 Angstrom break strength and emission line luminosity, indicating that departures from old elliptical galaxy continuum shapes are most likely due to non-stellar emission associated with the active nucleus.Comment: 14 pages, 6 figures, MNRAS in pres

    Radio-optical alignments in a low radio luminosity sample

    We present an optically-based study of the alignment between the radio axes and the optical major axes of eight z~0.7 radio galaxies in a 7C sample. The radio galaxies in this sample are ~20-times less radio luminous than 3C galaxies at the same redshift, and are significantly less radio-luminous than any other well-defined samples studied to date. Using Nordic Optical Telescope images taken in good seeing conditions at rest-frame wavelengths just longward of the 4000A break, we find a statistically significant alignment effect in the 7C sample. Furthermore, in two cases where the aligned components are well separated from the host we have been able to confirm spectroscopically that they are indeed at the same redshift as the radio galaxy. However, a quantitative analysis of the alignment in this sample and in a corresponding 3C sample from HST archival data indicates that the percentage of aligned flux may be lower and of smaller spatial scale in the 7C sample. Our study suggests that alignments on the 50-kpc scale are probably closely related to the radio luminosity, whereas those on the 15 kpc scale are not.     MicroRNA let-7c Is Downregulated in Prostate Cancer and Suppresses Prostate Cancer Growth

    Prostate cancer (PCa) is characterized by deregulated expression of several tumor suppressor or oncogenic miRNAs. The objective of this study was the identification and characterization of miR-let-7c as a potential tumor suppressor in PCa.Levels of expression of miR-let-7c were examined in human PCa cell lines and tissues using qRT-PCR and in situ hybridization. Let-7c was overexpressed or suppressed to assess the effects on the growth of human PCa cell lines. Lentiviral-mediated re-expression of let-7c was utilized to assess the effects on human PCa xenografts.We identified miR-let-7c as a potential tumor suppressor in PCa. Expression of let-7c is downregulated in castration-resistant prostate cancer (CRPC) cells. Overexpression of let-7c decreased while downregulation of let-7c increased cell proliferation, clonogenicity and anchorage-independent growth of PCa cells in vitro. Suppression of let-7c expression enhanced the ability of androgen-sensitive PCa cells to grow in androgen-deprived conditions in vitro. Reconstitution of Let-7c by lentiviral-mediated intratumoral delivery significantly reduced tumor burden in xenografts of human PCa cells. Furthermore, let-7c expression is downregulated in clinical PCa specimens compared to their matched benign tissues, while the expression of Lin28, a master regulator of let-7 miRNA processing, is upregulated in clinical PCa specimens.These results demonstrate that microRNA let-7c is downregulated in PCa and functions as a tumor suppressor, and is a potential therapeutic target for PCa

    Inhibition of let-7c Regulates Cardiac Regeneration after Cryoinjury in Adult Zebrafish

    The let-7c family of micro-RNAs (miRNAs) is expressed during embryonic development and plays an important role in cell differentiation. We have investigated the role of let-7c in heart regeneration after injury in adult zebrafish. let-7c antagomir or scramble injections were given at one day after cryoinjury (1 dpi). Tissue samples were collected at 7 dpi, 14 dpi and 28 dpi and cardiac function was assessed before cryoinjury, 1 dpi, 7 dpi, 14 dpi and 28 dpi. Inhibition of let-7c increased the rate of fibrinolysis, increased the number of proliferating cell nuclear antigen (PCNA) positive cardiomyocytes at 7 dpi and increased the expression of the epicardial marker raldh2 at 7 dpi. Additionally, cardiac function measured with echocardiography recovered slightly more rapidly after inhibition of let-7c. These results reveal a beneficial role of let-7c inhibition in adult zebrafish heart regeneration

    Dual Functions of Interferon Regulatory Factors 7C in Epstein-Barr Virus–Mediated Transformation of Human B Lymphocytes

    Epstein-Barr virus (EBV) infection is associated with several human malignancies. Interferon (IFN) regulatory factor 7 (IRF-7) has several splicing variants, and at least the major splicing variant (IRF-7A) has oncogenic potential and is associated with EBV transformation processes. IRF-7C is an alternative splicing variant with only the DNA-binding domain of IRF-7. Whether IRF-7C is present under physiological conditions and its functions in viral transformation are unknown. In this report, we prove the existence of IRF-7C protein and RNA in certain cells under physiological conditions, and find that high levels of IRF-7C are associated with EBV transformation of human primary B cells in vitro as well as EBV type III latency. EBV latent membrane protein 1 (LMP-1) stimulates IRF-7C expression in B lymphocytes. IRF-7C has oncogenic potential in rodent cells and partially restores the growth properties of EBV-transformed cells under a growth-inhibition condition. A tumor array experiment has identified six primary tumor specimens with high levels of IRF-7C protein—all of them are lymphomas. Furthermore, we show that the expression of IRF-7C is apparently closely associated with other IRF-7 splicing variants. IRF- 7C inhibits the function of IRF-7 in transcriptional regulation of IFN genes. These data suggest that EBV may use splicing variants of IRF-7 for its transformation process in two strategies: to use oncogenic properties of various IRF-7 splicing variants, but use one of its splicing variants (IRF-7C) to block the IFN-induction function of IRF-7 that is detrimental for viral transformation. The work provides a novel relation of host/virus interactions, and has expanded our knowledge about IRFs in EBV transformation

    Extremely red galaxy counterparts to 7C radio sources

    We present RIJHK imaging of seven radio galaxies from the 7C Redshift Survey (7CRS) which lack strong emission lines and we use these data to investigate their spectral energy distributions (SEDs) with models which constrain their redshifts. Six of these seven galaxies have extremely red colours (R-K>5.5) and we find that almost all of them lie in the redshift range 1<z<2. We also present near-infrared spectroscopy of these galaxies which demonstrate that their SEDs are not dominated by emission lines, although tentative lines, consistent with H-alpha at z=1.45 and z=1.61, are found in two objects. Although the red colours of the 7CRS galaxies can formally be explained by stellar populations which are either very old or young and heavily reddened, independent evidence favours the former hypothesis. At z~1.5 at least 1/4 of powerful radio jets are triggered in massive (>L*) galaxies which formed the bulk of their stars several Gyr earlier, that is at epochs corresponding to redshifts z>5. If a similar fraction of all z~1.5 radio galaxies are old, then extrapolation of the radio luminosity function shows that, depending on the radio source lifetimes, between 10-100% of the near-IR selected extremely red object (ERO) population undergo a radio outburst at epochs corresponding to 1<z<2. An ERO found serendipitously in the field of one of the 7CRS radio sources appears to be a radio-quiet analogue of the 7CRS EROs with an emission line likely to be [OII] at z=1.20. The implication is that some of the most massive elliptical galaxies formed the bulk of their stars at z>5 and these objects probably undergo at least two periods of AGN activity: one at high redshift during which the black hole forms and another one at an epoch corresponding to z~1.5.Comment: 20 pages, 10 figures, accepted for publication in MNRA

    The radio luminosity function of radio-loud quasars from the 7C Redshift Survey

    We present a complete sample of 24 radio-loud quasars (RLQs) from the new 7C Redshift Survey. Every quasar with a low-frequency (151 MHz) radio flux-density S_151 > 0.5 Jy in two regions of the sky covering 0.013 sr is included; 23 of these have sufficient extended flux to meet the selection criteria, 18 of these have steep radio spectra (hereafter denoted as SSQs). The key advantage of this sample over most samples of RLQs is the lack of an optical magnitude limit. By combining the 7C and 3CRR samples, we have investigated the properties of RLQs as a function of redshift z and radio luminosity L_151. We derive the radio luminosity function (RLF) of RLQs and find that the data are well fitted by a single power-law with slope alpha_1=1.9. We find that there must be a break in the RLQ RLF at log_10(L_151 / W Hz^-1 sr^-1) < 27, in order for the models to be consistent with the 7C and 6C source counts. The z-dependence of the RLF follows a one-tailed gaussian which peaks at z=1.7. We find no evidence for a decline in the co-moving space density of RLQs at higher redshifts. A positive correlation between the radio and optical luminosities of SSQs is observed, confirming a result of Serjeant et al. (1998). We are able to rule out this correlation being due to selection effects or biases in our combined sample. The radio-optical correlation and best-fit model RLF enable us to estimate the distribution of optical magnitudes of quasars in samples selected at low radio frequencies. We conclude that for samples with S_151 < 1 Jy one must use optical data significantly deeper than the POSS-I limit (R approx 20), in order to avoid severe incompleteness.Comment: 28 pages with 13 figures.     A complete sample of quasars from the 7C redshift survey

    We present details of a new sample of radio-loud quasars drawn from 0.013 sr of the 7C Redshift Survey. This sample is small (21 quasars) but complete in that every object with an unresolved nucleus and/or broad emission lines with S(151MHz) > 0.5 Jy has been discovered. The dependence of the quasar fraction with redshift and radio luminosity is investigated, providing new evidence supporting the unification of radio-loud quasars and powerful radio galaxies. This 7C sample is compared with optically-selected quasars, in order to determine whether there are systematic biases in the different selection techniques. There are no lightly reddened (Av approx. 1) quasars in our sample amongst the 14 with z < 2. The discovery of a reddened quasar at z = 2.034 and its implications are discussed.     Let-7c down-regulation in Helicobacter pylori-related gastric carcinogenesis

    open12siAberrant let-7c microRNA (miRNA) expression has been observed in Helicobacter pylori-related gastric cancer (GC) but fragmentary information is available on the let-7c dysregulation occurring with each phenotypic change involved in gastric carcinogenesis. Let-7c expression was assessed (qRT-PCR) in a series of 175 gastric biopsy samples representative of the whole spectrum of phenotypic changes involved in H. pylori-related gastric oncogenesis including: i) normal gastric mucosa, as obtained from dyspeptic controls (40 biopsy samples); ii) non-atrophic gastritis (40 samples); iii) atrophic-metaplastic gastritis (35 samples); iv) intra-epithelial neoplasia (30 samples); v) GC (30 samples). Let-7c expression was also tested in 20 biopsy samples obtained from 10 patients before and after H. pylori eradication therapy (median follow-up: 10 weeks; range: 7-14). The results obtained were further validated by in situ hybridization on multiple tissue specimens obtained from 5 surgically treated H. pylori-related GCs. The study also included 40 oxyntic biopsy samples obtained from serologically/histologically confirmed autoimmune gastritis (AIG: 20 corpus-restricted, non-atrophic; 20 corpus-restricted, atrophic-metaplastic). Let-7c expression dropped from non-atrophic gastritis to atrophic-metaplastic gastritis, intra-epithelial neoplasia, and invasive GC (p<0.001). It rose again significantly following H. pylori eradication (p=0.009). As in the H. pylori model, AIG also featured a significant let-7c down-regulation (p<0.001). The earliest phases of the two pathways to gastric oncogenesis (H. pylori-environmental and autoimmune host-related) are characterized by similar let-7c dysregulations.
