KONSERVASI TANAMAN ANGGREK Gramatophyllum SECARA IN VITRO MELALUI PERTUMBUHAN MINIMAL MENGGUNAKAN PACLOBUTRAZOL

Abstract

<p>Tujuan penelitian ini adalah mengkaji medium pertumbuhan minimal bagi <em>plantlet</em> anggrek <em>Gramatophyllum</em> dan pertumbuhan kembali <em>plantlet</em> setelah mengalami pertumbuhan minimal selama empat bulan. Penelitian dilakukan secara eksperimental menggunakan rancangan acak lengkap. Variabel bebas dalam penelitian ini adalah penambahan paclobutrazol ke dalam medium tanam dengan konsentrasi berturut-turut: 1, 3, dan 5 mg/l. Variabel terikatnya adalah pertumbuhan tunas, dengan parameter pengamatan jumlah daun dan panjang tunas setelah dipelihara dalam medium dengan penambahan paclobutrazol dengan berbagai konsentrasi selama empat bulan. Hasil penelitian menunjukkan bahwa pertumbuhan anggrek pada medium dengan penambahan paclobutrazol pada semua konsentrasi perlakuan (1, 3, dan 5 mg/l) mengalami hambatan, baik dalam jumlah daun maupun panjang tunas. Hambatan pertumbuhan yang terjadi bervariasi berdasar konsentrasi paclobutrazol yang ditambahkan. Perlakuan paclobutrazol pada <em>plantlet</em> <em>Grammatophyllum</em> dapat menurunkan panjang tunas hingga 33,3% bila dibandingkan dengan kontrol yaitu pada konsentrasi 1 mg/l. Jumlah daun juga mereduksi hingga 29% bila dibandingkan dengan kontrol pada konsentrasi paclobutrazol 5 mg/l. Hasil tersebut menunjukkan bahwa paclobutrazol dapat digunakan dalam konservasi jangka pendek pada <em>plantlet</em> <em>Grammatophyllum</em> pada konsentrasi 1-5 mg/l. Konservasi <em>Grammatophyllum</em> menggunakan paclobutrazol hendaknya tidak melebihi empat bulan karena akan menurunkan kualitas <em>plantlet</em>.</p><p> </p><p>The objective of this research was to study the minimum growth medium for Grammatophylum plantlet and the plantlets regeneration after grown in paclobutrazol medium at least in four months. The study was conducted experimentally using a complete random design. The independent variable in this study was concentrations of paclobutrazol added into the planting medium : 1, 3, and 5 mg/l. The dependent variable was the growth of shoot and the observed parameter of shoot growth was the number of leaves and shoot length after maintained in the medium with the addition of paclobutrazol in various concentrations for four months. The results showed that the growth of orchids on the medium with the addition of paclobutrazol treatment at all concentrations (1, 3, and 5 mg/l) had growth inhibition both in the number of leaves and shoot length. Growth inhibition that occured was varied depend on the addition of paclobutrazol concentration. Paclobutrazol treatment on Grammatophyllum plantlets was reduced the shoot length until 33.3% compared to the control at concentration of 1 mg/l. The number of leaves was also reduced until 29% compared to the control group at concentration of 5 mg/l paclobutrazol. These results indicated that paclobutrazol could be used as the short-term conservation of Grammatophyllum plantlets at 1-5 mg/l concentration. The conservation of Grammatophyllum using paclobutrazol should not exceed more than four months because it will reduce the quality of plantlets.</p
