THE IMPACT OF DIFFERENT EXTENDERS ON QUALITATIVE TRAITS OF A BULL SEMEN

Abstract

Namen diplomskega dela je določiti tisti razredčevalec, ki daje boljše rezultate v smislu kvalitativnih lastnosti bikovega semena (gibljivost, patološka gibljivost, negibljivost, progresivna gibljivost). Ejakulate smo odvzemali 10 bikom svetlo lisaste pasme, ki so bili stari od 3 do 6 let, in sicer po 2 ejakulata, ki sta kasneje tvorila en vzorec. Ejakulate smo redčili z dvema razredčevalcema, in sicer s Triladylom, ki vsebuje jajčni rumenjak, in Bioxcellom, ki ne vsebuje beljakovin živalskega izvora. Označene vzorce smo napolnili v slamice in jih ohlajali 3 in 24 ur. Sledila je globoka zamrznitev na –196 °C. Računalniška analiza semena je potekala s pomočjo sperma-analizatorja Hamillton Thorn- IVOS10, takoj po odtajanju na 38 °C ter po 24-tih urah na 4 °C (Müllerjev rezistenčni test). Najboljše rezultate smo dobili pri 3-urni ekvilibraciji v razredčevalcu Bioxcell. V našem primeru bi tako lahko sklepali, da je razredčevalec Bioxcell boljša izbira kot Triladyl in da je 3-urna ekvilibracija boljša od 24-urne.The aim of the study was to determine the extender that gives better results in relation to qualitative characteristics of the bull\u27s sperm cell (motility, pathological motility, immotility, progressive motility). Ejaculates have been taken from ten Simmental bulls in the age of 3 to 6 years2 ejaculates from each bull that later formed one sample. Ejaculates have been diluted with two extenders, with Triladyl containing the yolk and Bioxcell which did not contain any animal protein. Straws were filled with samples and cooled down for three and twenty-four hours, respectively. Samples were deep frozen to -196 °C. The computer analysis of the sperm cell took place with the help of sperm-analyser Hamillton Thorn – IVOS10 right after the thawing at 38°C and after 24 hours on 4°C (Muller\u27s resistance test). The best results were obtained after 3 hours of equilibration time in Bioxcell. On the basic of results, we can assume that the Bioxcell extender is a better choice than Triladyl, and that a 3 hour equilibration time is better than a 24-hours one
