Micropropagation and Mutation of Dieffenbachia maculata 'Rudolph Roehrs-86M1'

Abstract

乳羅黛粉葉誘變種'Rudolph Roehrs-86M1'以幼嫩完全展開葉切成 l × 1cm 方塊為培殖體,培養於含 1/2MS、蔗糖 20g/l、洋菜 6g/l、BA 5mg/l及NAA O.5mg/l之培養基,可由葉片誘導較多的癒傷組織。將長寬5 × 5mm之癒傷組織塊培養於1/2MS、蔗糖20g/l、洋菜 6g/l、BA 5mg/l以及NAA0.125mg/l之培養基經5-6週,可再分化植株。微體插穗長度在2.5公分以上者,於基部以NAA 0.1%粉劑處理後,瓶外發根率最佳。The young unfolded leaf of Dieffenbachia maculate ‘Rudolph Roehrs-86M1' was cut into 1 × 1 cm pieces. Then explants cultured on the medium containing 1/2 MS, sugar 20g/l, agar 6g/l, BA 5 mg/l and NAA 0.5g/l. grew more callus.\ud Shoot redifferentiated from callus explants of 5mm × 5mm cultured on the medium containing 1/2 MS, agar 6g/l, sugar 20g/l, BA 5 mg/l and NAA 0.125 mg/l. Shoot longer than 2.5 cm rooted well ex vitro when shoot base was coated with NAA 0.1%
