插花用海綿在移植前培養之應用

Abstract

為健化瓶苗,改善長途運輸後之品質,簡化出瓶移植時的操作,並提高出瓶成活率。將非洲菊(Gerbera jamesonii H. Bolus ex Hook. f.)瓶苗以插花用海綿(oasis)為液體培養時之支撐物,期能代替傳統的洋菜培養基。培養液使用半量之MS基本培養配方,再添加IBA 1mg/l、蔗糖 15g/l的配方,pH值調整為5.7。培養5週後,因pH值僅2.9,培殖體生長勢差,瓶苗存活率低。僅13.9%存活。為改善其培養效果,以培養前利用1N之NaOH調整培養液pH植為5.0,5.5,6.0,6.5,7.0後進行培養5週,結果沒有預期提高pH值的效果,也無法提高瓶苗成活率。但使用經泡水0,2,4,6,8,10天處理之插花用海綿進行培養,結果隨泡水天數增加,瓶苗存活率可由13.9%提高至53%,生長勢也明顯改善。For hardening the plantlets, improving transportation quality, simplifying process from ex vitro to in vivo, and increasing the survival rate, explants of Gerbera jamesonii H. Bolus ex Hook. f. were cultivated on oasis absorbing. The medium of 1/2 MS contained IBA 0.1 mg/l and sucrose 15g/l, and the were adjusted to pH 5.7 in vitro. Explants cultivated on oasis were low survival grew very poor. Adjusted pH value by 1N NaOH did not improve survival rate og explants from in vitro to ex vitro. Moreover, the soaking effect increased by extending the soaking days and change times of soaking water
