Abstract

This dissertation describes the recovery of a steroid (6#alpha#-methyl-prednisolone) biotransformed by Arthrobacter simplex cells. An integrated process that combined cell growing and harvesting with steroid biotransformation and recovery was used. Commercial cross-flow and vorticular flow (rotary membrane) filtration modules were used for the separation. Several membranes for ultra and microfiltration were tested. The membrane filtration was run in concentration and diafiltration modes of operation. The permeation fluxes and the steroid transmissions through the membrane were analyzed and the membrane fouling by steroid was quantified and discussed. among the filtration systems studied, the vorticular flow filtration system, with a hydrophilic membrane, proved to be the most adequate to obtain purified steroid solutions. Studies of permeation flux modelling were conducted for ultra and microfiltration with the vorticular flow filtration system. The module was submitted to filtration runs with standardized biotransformation media in a total recirculation mode of operation. The models of the resistances in series and the mass transfer model were both used, each of which applied to different regions of transmenbrane pressure.
