SORPTION METHODS FOR EXTRACTION AND CLEANING OF PANCREATIC ENZYMES: RIBONUCLEASE, DEOXYRIBONUCLEASE, TRYPSIN AND CHYMOTRYPSIN

Abstract

The work covers the pancreatic ribonuclease, deoxyribonuclease, trypsin, chymotrypsin and also ionogenic and non-ionogenic sorbents. The aim of the work is to study the regularities in the molecular and ion-exchange sorption of pancreatic enzymes and to develop the sorption chromatographic extraction and cleaning process technology on their base. The following methods have been used: spectrophotometric method, Loury method, medified Kunitz method, method of limited volume (kinetic one), thermodynamic analysis method, statistical methods, regression analysis. The equilibrium, kinetic and dynamic characteristics and thermodynamic parameters of sorption in the pancreatic enzymes by different sorbents have been studied; the intercommunication in structure of the sorbents has been determined, and their influence of the selectivity and capacity of the sorption has been proved. The procedure of making enzymes of the pancreas has been developed. The extraction and cleaning of pancreatic enzyme complex of the non-tranparent colloidal solutions on the sulphocationic exchanger KU-23 in the ion-exchange columns have been performed. The scaling of sorption process has been performed. The production process of pancreatic enzymes has been developed. The ion-exchange technology of extracting and cleaning pancreatic enzymes has been put into the production. The process time has been reduced 5 times, the yield of finished products has been increased by 30%, the quality has been improved.
