Common bean breeding to angular leaf spot resistance: validation of SCAR molecular markers, genes pyramiding, allelisms tests and characterization of elite lines

Abstract

Neste trabalho são apresentados resultados de pesquisas desenvolvidas no âmbito do Programa de Melhoramento do Feijoeiro da Universidade Federal de Viçosa (UFV), o qual tem visado o melhoramento genético para resistência à mancha-angular (Pseudocercospora griseola), antracnose (Colletotrichum lindemuthianum) e ferrugem (Uromyces appendiculatus). Inicialmente, foram validados os marcadores moleculares SCAR SE04640a, SAA07950a e SAO12950a como sendo ligados a genes de resistência à mancha-angular. Paralelamente, foram selecionadas e caracterizadas linhagens e populações de feijão carioca com resistência a doenças e outras características agronômicas favoráveis. Neste caso, foram realizadas seleções assistidas por marcadores moleculares ligados a genes de resistência às três doenças, ensaios a campo, e caracterizações fenotípicas com diversas raças dos três patógenos. Também foram realizados testes de alelismo para o loco de resistência à mancha-angular, presente no cultivar Ouro Negro , frente a outros genótipos já utilizados pelo referido Programa. Finalmente, foram caracterizados importantes cultivares e linhagens elite quanto à reação a diversas raças de Pseudocercospora griseola oriundos do estado de Minas Gerais.This work present research data developed in the common bean breeding program of the Universidade Federal de Viçosa (UFV), whose main target is genetically breeding common bean aiming at pyramiding resistance to angular leaf spot (Pseudocercospora griseola), anthracnose (Colletotrichum lindemuthianum) and rust (Uromyces appendiculatus) diseases. Initially, the SCAR molecular markers SE04640a, SAA07950a and SAO12950a were validated as linked to resistance genes to angular leaf spot. At the same time, lines and populations of the "carioca" common bean were selected and characterized with respect to resistance to diseases and other favorable agronomic traits. In this case, assisted selections were carried out by molecular markers linked to the three diseases resistance genes, as well as field essays and phenotypic characterizations with several races of the three pathogens. Allelism tests were also carried out for the locus of resistance to the angular leaf spot, present in the cultivar 'Ouro Negro', genotypes already used by the mentioned breeding. Program.
