RESPONSE OF PLATELETS TO SEVERAL THROMBOTIC AGONISTS

Abstract

THE RESPONSE OF HUMAN AND RAT PLATELETS TO THROMBIN, EPINEPHRINE AND FLUORIDE IONS WAS INVESTIGATED IN THIS PHD THESIS. IT WAS OF INTEREST TO STUDY THE MECHANISM(S) THAT THESE AGONISTS AFFECT THE LEVELS OF INTRACELLULAR FREE CALCIUM ([CA]I), SODIUM ([NA+]I) AND HYDROGEN IONS (PHI) CONCENTRATION. BASAL [NA]I LEVELS IN HUMAN PLATELETS WERE FOUND AP. 5MM, WHILE STIMULATION BY THROMBIN EVOKES A DOSE-RESPONSE RISE, THE MAIN PART OF WHICH (50%) IS ATTRIBUTABLE TO NA+ INFLUX AND THE REST TO OTHER BIOCHEMICAL PATHWAYS. EPINEPHRINE FAILED TO AFFECT [NA]I, BUT IT WAS FOUND THAT THE PRESENCE OF EXTRACELLULAR NA+ ARE NECESSARY FOR FULL PLATELET ACTIVATION. FLUORIDE IONS FAIL TO ACTIVATE NA+/H+, AND THEIR EFFECT IS PARTLY INHIBITED BY SUBSTANCES WHICH ELEVATE CAMP IN PLATELETS BASAL [NA+]I IN RAT PLATELETS WERE FOUND STIMES HIGHER THAN HUMAN PLATELETS. THE EFFECT OF THE ABOVE 3 AGONIST WAS INVESTIGATED IN RAT PLATELETS.
