ISOLATION OF A NUCLEAR RNP-COMPLEX FROM RAT LIVER. MORPHOLOGICAL- BIOCHEMICAL AND IMMUNOLOGICAL STUDY

Abstract

A NOVEL RNP-COMPLEX HAS BEEN ISOLATED FROM RAT LIVER NUCLEI. IT IS HIGHLY ENRICHED IN STRUCTURES WITH MEAN DIAMETERS 200-250 A, RESISTANT TO THE ACTION OF NUCLEASE. THESE STRUCTURES SEDIMENT ON A SUCROSE GRADIENT AS >80 S COMPONENTS, AND THEY CONTAIN MAINLY BASIC PROTEINS OF MW=130, 115, 78, 76 KD AS WELL AS THE KNOWN U-SNRNA SPECIES (U1, U2, U4, U5, U6). THE STRUCTURES BAND AT P=1,3650 IN ISOPYARIC CSCL GRADIENT. THE ABOVE CHARACTERISTICS OF THIS RNP-COMPLEX TAKEN TOGETHER WITH RECENT FINDINGS IN THE LITERATURE, SUGGEST A POSSIBLE RELATIONSHIP BETWEEN INTERCHROMATIN GRANULES AND THE > 80 S STRUCTURES. FOLLOWING IMMUNIZATION OF RABBITS WITH THESE > 80 S STRUCTURES, WE HAVE OBTAINED A SERUM (SERUM S2), WHICH SHOWS SPECIES SPECIFICITY AND REACTS MAINLY WITH NUCLEOLI. THESERUM ALSO GIVES A SPECKLED PATTERN IN THE NUCLEOPLASM. THE MAJOR ANTIBODY OFSERUM S2 (ANTIBODY B), THAT RECOGNIZES A PROTEIN OF MW=94 KD, REACTS WITH THENUCLEOLI, BUT IT ALSO GIVES A SPECKLED PATTERN AFTER DRB TREATMENT OF THE CELLS. THIS LATTER OBSERVATION AND RECENT PUBLISHED DATA, WHICH CONNECT INTERCHROMATION GRANULES WITH NUCLEOLI, SUPPORT THE VIEW THAT THESE > 80 S RNP-STRUCTURES ARE INDEED THE INTERCHROMATION GRANULES OF THE NUCLEUS.
