The Enclisis on subordinate clauses in the history of European Portuguese

A colocação pronominal é um dos assuntos mais estudados da língua portuguesa, mas sempre priorizando as orações principais finitas onde há grande variação na colocação dos clíticos, com a mudança gramatical ocorrida no século 18 (Galves, Brito e Paixão de Sousa 2005, Galves, Namiuti e Paixão de Sousa2005; Paixão de Sousa, 2004). As orações dependentes sempre foram consideradas como contexto de próclise categórica. No entanto, nos dados extraídos do Corpus Histórico Tycho Brahe, apesar da predominância proclítica, foram atestados dados de ênclise em dependentes. A ênclise ocorre em orações dependentes nas gramáticas do Português - Clássico e Europeu moderno- sempre que há pelo menos um constituinte entre o complementador e o verbo. A ênclise ocorre em todo o período - Português Clássico ao Português Europeu moderno - em vários tipos de oração: relativas, completivas, dependentes iniciadas em porque. Mostro ainda que há uma estabilidade ao longo do tempo na colocação de clíticos em orações dependentes, ao contrário do que acontece nas orações principais A pergunta a ser respondida foi: como estas duas gramáticas geram a ênclise nas dependentes? Analiso o fenômeno com base no CP expandido de Rizzi (1997), e ainda retomando a proposta de Galves e Sandalo 2009 para a colocação de clíticos no PCl e no PEAbstract: The aim of this work is to investigate the enclisis on subordinate clauses in the history of the European Portuguese. The clitic placement is one of the most popular subjects about the Portuguese language, but these researches always focus on the main clauses, there is a large variation on the clitics placement, after the language change around 18 century (Galves, Brito e Paixão de Sousa 2005; Paixão de Sousa 2004). The subordinate clauses were always considered as categorical proclisis context. Nevertheless, despite the high rates of proclisis on subordinates clauses, there are enclisis data from Tyhco Brahe Parsed Corpus of historical Porutuguese. The enclisis is only possible on subordinate clauses in Classical Portuguese and European Portuguese, when there is at least on constituent between the complement and the verb. The enclisis occurs in all type of subordinate clauses: relatives, completives, dependent clauses beginning with porque (because), during all the period - from Classical Portuguese to European Portuguese, and there are a stability during time. PI3K inhibition synergizes with glucocorticoids but antagonizes with methotrexate in T-cell acute lymphoblastic leukemia
The PI3K pathway is frequently hyperactivated in primary T-cell acute lymphoblastic leukemia (T-ALL) cells. Activation of the PI3K pathway has been suggested as one mechanism of glucocorticoid resistance in T-ALL, and patients harboring mutations in the PI3K negative regulator PTEN may be at increased risk of induction failure and relapse. By gene expression microarray analysis of T-ALL cells treated with the PI3K inhibitor AS605240, we identified Myc as a prominent downstream target of the PI3K pathway. A significant association was found between the AS605240 gene expression signature and that of glucocorticoid resistance and relapse in T-ALL. AS605240 showed anti-leukemic activity and strong synergism with glucocorticoids both in vitro and in a NOD/SCID xenograft model of T-ALL. In contrast, PI3K inhibition showed antagonism with methotrexate and daunorubicin, drugs that preferentially target dividing cells. This antagonistic interaction, however, could be circumvented by the use of correct drug scheduling schemes. Chronic activation of γ2 AMPK induces obesity and reduces β cell function
Despite significant advances in our understanding of the biology determining systemic energy homeostasis, the treatment of obesity remains a medical challenge. Activation of AMP-activated protein kinase (AMPK) has been proposed as an attractive strategy for the treatment of obesity and its complications. AMPK is a conserved, ubiquitously expressed, heterotrimeric serine/threonine kinase whose short-term activation has multiple beneficial metabolic effects. Whether these translate into long-term benefits for obesity and its complications is unknown. Here, we observe that mice with chronic AMPK activation, resulting from mutation of the AMPK γ2 subunit, exhibit ghrelin signaling-dependent hyperphagia, obesity, and impaired pancreatic islet insulin secretion. Humans bearing the homologous mutation manifest a congruent phenotype. Our studies highlight that long-term AMPK activation throughout all tissues can have adverse metabolic consequences, with implications for pharmacological strategies seeking to chronically activate AMPK systemically to treat metabolic disease23582183

Orientador: José Eduardo Ribeiro de PaivaDissertação (mestrado) - Universidade Estadual de Campinas, Instituto de ArtesResumo: Este trabalho trata da tecnologia aplicada em produções de áudio em estúdios com corais mistos, polifônicos, amadores e adultos, tendo a cidade de São Paulo e cidades adjacentes como referências. Apresenta como objetivo central prover material de pesquisa capaz de auxiliar o regente coral em performances no estúdio, entendendo qual deve ser a capacitação deste para esta função dentro do ambiente controlado de gravação e, como objetivo secundário, apurar as etapas para a produção de um registro de áudio em estúdio. Trata-se de um trabalho direcionado a regentes que atuam à frente de coros mistos, polifônicos e adultos de natureza amadora (SATB) que pretendem submeter seu coro a uma performance de música gravada. Após uma reflexão introdutória, são apresentados o papel do coro e a função do regente, etapas do processo de gravação, bem como um levantamento acerca dos avanços tecnológicos no processo de captação e edição de áudio, analisando seus desdobramentos na relação entre regentes, coralistas e ouvintes. Na sequência, há o material compilado dos corais que, ao longo de sua carreira, registraram em áudio momentos da sua produção artística e a transcrição dos depoimentos recolhidos durante a execução deste projeto, findando com a análise do material coletadoAbstract: This work deals with the technology applied in audio productions in mixed coral, polyphonic, amateur and adult studios, with the city of São Paulo and adjacent cities as references. Oropouche virus infection and pathogenesis are restricted by MAVS, IRF-3, IRF-7, and type i interferon signaling pathways in nonmyeloid cells
Oropouche virus (OROV) is a member of the Orthobunyavirus genus in the Bunyaviridae family and a prominent cause of insect-transmitted viral disease in Central and South America. Despite its clinical relevance, little is known about OROV pathogenesis. To define the host defense pathways that control OROV infection and disease, we evaluated OROV pathogenesis and immune responses in primary cells and mice that were deficient in the RIG-I-like receptor signaling pathway (MDA5, RIG-I, or MAVS), downstream regulatory transcription factors (IRF-3 or IRF-7), beta interferon (IFN-β), or the receptor for type I IFN signaling (IFNAR). OROV replicated to higher levels in primary fibroblasts and dendritic cells lacking MAVS signaling, the transcription factors IRF-3 and IRF-7, or IFNAR than in wild-type (WT) cells. In mice, deletion of IFNAR, MAVS, or IRF-3 and IRF-7 resulted in uncontrolled OROV replication, hypercytokinemia, extensive liver damage, and death, whereas WT congenic animals failed to develop disease. Unexpectedly, mice with a selective deletion of IFNAR on myeloid cells (CD11c Cre+ Ifnar f/f or LysM Cre+ Ifnar f/f) did not sustain enhanced disease with OROV or a selective (flox/flox) deletion La Crosse virus, a closely related encephalitic orthobunyavirus. In bone marrow chimera studies, recipient irradiated Ifnar −/− mice reconstituted with WT hematopoietic cells sustained high levels of OROV replication and liver damage, whereas WT mice reconstituted with Ifnar −/− bone marrow were resistant to disease. Estimating species diversity and distribution in the era of Big Data: to what extent can we trust public databases?
Massive digitalization of natural history collections is now leading to a steep accumulation of publicly available species distribution data. However, taxonomic errors and geographical uncertainty of species occurrence records are now acknowledged by the scientific community - putting into question to what extent such data can be used to unveil correct patterns of biodiversity and distribution. We explore this question through quantitative and qualitative analyses of uncleaned versus manually verified datasets of species distribution records across different spatial scales. The American tropics. As test case we used the plant tribe Cinchoneae (Rubiaceae). We compiled four datasets of species occurrences: one created manually and verified through classical taxonomic work, and the rest derived from GBIF under different cleaning and filling schemes. We used new bioinformatic tools to code species into grids, ecoregions, and biomes following WWF's classification. We analysed species richness and altitudinal ranges of the species. Altitudinal ranges for species and genera were correctly inferred even without manual data cleaning and filling. However, erroneous records affected spatial patterns of species richness. They led to an overestimation of species richness in certain areas outside the centres of diversity in the clade. The location of many of these areas comprised the geographical midpoint of countries and political subdivisions, assigned long after the specimens had been collected.Open databases and integrative bioinformatic tools allow a rapid approximation of large-scale patterns of biodiversity across space and altitudinal ranges. We found that geographic inaccuracy affects diversity patterns more than taxonomic uncertainties, often leading to false positives, i.e. overestimating species richness in relatively species poor regions. Public databases for species distribution are valuable and should be more explored, but under scrutiny and validation by taxonomic experts.
