    Role of neurofascin in gephyrin cluster formation and localization during inhibitory synaptogenesis in the central nervous system

    Membrangebundene Zelladhäsionsmoleküle (CAMs) stellen Schlüsselkomponenten bei der Entstehung exzitatorischer Synapsen im ZNS dar. Bislang war wenig zu deren Rolle bei der Entstehung inhibitorischer Synapsen bekannt. Insbesondere der funktionelle Zusammenhang zwischen CAMs und der Clusterbildung des postsynaptischen Gerüstproteins Gephyrin, das die Akkumulation von GABAA-Rezeptoren an inhibitorischen Postsynapsen steuert, muss noch aufgeklärt werden. Neurofascin gehört zur L1-Unterfamilie der Immunglobulin-ähnlichen CAMs (IgCAMs). In vivo wurde gezeigt, dass es die Entstehung und Lokalisation von GABAergem Input an zerebellären Purkinje-Neuronen reguliert. Neurofascin stellt deshalb ein aussichtsvolles Kandidatenmolekül für die Rekrutierung von Gephyrin an inhibitorische Postsynapsen dar. Zu frühen Entwicklungszeitpunkten korrelierten die Bildung von Gephyrinclustern und ihre Translokation an den Axonhügel mit der Expression von Neurofascin auf dem Soma und Axoninitialsegment (AIS) dissoziierter hippokampaler Neurone. Des Weiteren wurde zu frühen Zeitpunkten der Synaptogenese hauptsächlich eine Neurofascin-Isoform exprimiert, der die fünfte Fibronektin-III ähnliche Domäne fehlt (NF-5te FN-III). Im Gegensatz dazu wurde die Isoform NF+5te FN-III erst im adulten Rattengehirn exprimiert. Die Transfektion von Expressionsvektoren unterschiedlicher Neurofascin-Isoformen bzw. Deletionsmutanten ergab, dass die embyronale Isoform NF-5te FN-III notwendig für die Gephyrinclusterbildung ist. Dieser Prozess scheint von extrazellulären Interaktionspartnern auf prä;- bzw. postsynaptischen Komponenten abhängig zu sein. Gegenwärtig ist die Identität eines Neurofascin-Interaktionspartners jedoch unbekannt. Punktmutationen in der zytoplasmatischen Domäne Neurofascins deuten darauf hin, dass dieses durch Aktivierung intrazellulärer Signalkaskaden die Clusterbildung von Gephyrin induziert. Mittels shRNA-vermitteltem Knockdown wurde gezeigt, dass Neurofascin notwendig für die Translokation von Gephyrin an den Axonhügel hippokampaler Neurone ist. Überdies ist Neurofascin sogar hinreichend für die funktionelle Wiederherstellung anomal lokalisierter Gephyrincluster nach Neurofascin-Knockdown. Die Expression von NF-5te FN-III resultierte in einer Akkumulation exogen exprimierten GFP-Gephyrins. Dies deutet auf einen funktionellen Zusammenhang zwischen Neurofascin und Gephyrin hin. Kolokalisationsstudien in HEK293- bzw. PC12E2-Zellen lieferten jedoch keinen Hinweis für eine direkte Neurofascin-Gephyrin-Interaktion, die zu den beobachteten Effekten führt.Membrane-bound cell adhesion molecules (CAMs) were discovered to be key players in initiating excitatory synapse formation in the CNS. However, so far little is known about the role of CAMs in inhibitory synapse development. In particular, a functional link between CAMs and the clustering of postsynaptic scaffold component gephyrin, which is a critical determinant of y-aminobutyric acid A (GABAA) clustering, still needs to be elaborated. Neurofascin belongs to the L1-subgroup of the immunoglobuline like CAMs (IgCAMs). In vivo Neurofascin has been shown to direct the formation and localization of GABAergic Input on cerebellar Purkinje neurons. Thus it serves as a candidate molecule recruiting gephyrin to inhibitory postsynaptic sites. At early stages of inhibitory synaptogenesis, formation of gephyrin clusters and their translocation to the axon hillock correlated with a somatic expression of neurofascin in rat hippocampal neurons. Furthermore, a neurofascin splice variant lacking the extracellular fifth fibronectine-III like domain (NF-5te FN-III) was predominantely expressed at early stages of synapse formation. In contrast expression of NF+5te FN-III harbouring the fifth fibronectine-III like domain was prominent only in adult brain. Transfection of expression vectors for different splice variants and deletion mutants of neurofascin revealed that the embryonic neurofascin isoform NF-5te FN-III is required for the formation of gephyrin clusters. This process is presumably dependent on extracellular interactions with molecules on pre- and postsynaptic terminals, respectively. However, possible interaction partners for neurofascin are unknown. Point mutations in the cytoplasmatic domain of neurofascin inhibited the formation of gephyrin clusters suggesting intracellular signal transduction pathways triggered by neurofascin. Furthermore, expression of neurofascin is necessary for the translocation of gephyrin clusters to the axon hillock of hippocampal neurons as shown by shRNA-mediated knockdown. In addition, overexpression of an embryonic neurofascin isoform was sufficient for functional rescue after knockdown of endogenous neurofascin. Expression of NF-5te FN-III resulted in the accumulation of exogenous GFP-Gephyrin at the axon initial segment AIS suggesting a functional link between neurofascin and gephyrin. However, colocalization studies in HEK293 and PC12E2 cells, respectively, did not provide an indication of direct neurofascin gephyrin interactions leading to the observed effects

    Biological control of Striga hermonthica (Del.) Benth. using formulated mycoherbicides under Sudan field conditions

    Striga hermonthica is a parasitic flowering plant belonging to the family Orobanchaceae. It is a root parasite that attacks sorghum, maize, millet and several grass weeds in the semi-arid Tropics. In Sudan, Striga is widespread in irrigated and rainfed areas and considered the main biotic constraint in production of sorghum, the main staple food for the majority of Sudanese people. More than 500,000 hectares under rainfed cultivation are heavily infested with Striga, which commonly results in significant yield losses of 70 ? 100%. It has become obvious that there is no simple, fast and inexpensive solution to the Striga problem in Africa. Biological control is considered a potential cost-effective and environmentally safe means for reducing weed populations in crops, forests, or rangelands where low profit margins prevent large herbicide expenditure. Biological control using microorganisms (especially phytopathogenic fungi) showed a high efficacy in controlling S. hermonthica under controlled and field conditions. However, so far it did not come to practical field application. This could be attributed to environmental obstacles or due to the lack of appropriate delivery systems. The pathogenicity of two fungal isolates indigenous to Sudan (Fusarium nygamai [FN] and F. ?Abuharaz? [FA] isolate) against Striga has been studied using infected sorghum grains or a spore suspension as inoculum. These formulations were very effective in controlling Striga under controlled and natural conditions; however, a high level of fungal inoculum (approximately 800 kg ha-1 for the grain inoculum) would be required for effective control, which arises a lot of problems e.g. concerning sterilization and transportation. Such problems can be overcome by adopting an appropriate formulation technology. Granular formulations such as ?Pesta? and alginate pellets were found to be suitable delivery systems for controlling weeds. ?Pesta? granules are made by encapsulating bioagents in a gluten matrix. Alginate formulations are prepared by incorporating the biocontrol agent?s propagules in a sodium alginate solution, which is dripped to a calcium chloride or calcium gluconate solution. Alginate pellets are then formed by ionotrophic gelation. The main objectives of this study were: (a) to study the efficacy of the two Fusarium species in controlling Striga under field conditions using ?Pesta? and alginate formulations, (b) evaluate the effect on sorghum yield, (c) determine the optimum dose of the formulated material, (d) investigate the persistence of the formulated fungal isolates in the soil, and (e) study the efficacy of seed treatments as an alternative delivery system. Furthermore, for environmental safety reasons the newly isolated F. ?Abuharaz? isolate was tested for its ability to produce some of the most important mycotoxins. Harvested sorghum seeds out of the fungus-treated plots were also investigated for their mycotoxins content. A prerequisite to be able to formulate biocontrol fungi is the development of an inexpensive method of inoculum production that yields sufficient biomass containing viable, highly virulent propagules. Chlamydospores are the soil-persisting propagules of many Fusarium species and considered as ideal propagules to be used in granular formulations. For this reason, finding a medium suitable for the production of chlamydospores by the two Fusarium isolates was one of the specific objectives of this study. Different media were tested among them Special Nutrient-poor Broth (SNB) + yeast gave the highest number of chlamydospores (105 ml-1) in both isolates throughout the incubation period. However, both isolates generally did not form sufficient chlamydospores to be used within a bioherbicide formulation. Richard?s solution gave the highest number of microconidia (108 ml-1) after five days of incubation and hence it was selected as growth medium for formulation purposes throughout this study. FN and FA were successfully formulated in ?Pesta? and alginate granules amended either with 10% wheat flour or 6% sorghum flour or yeast extract. Alginate granules generally gave higher numbers of colony forming units (cfu) per g of formulated material compared to ?Pesta?. Alginate preparations amended with 6% sorghum flour or yeast extract had significantly higher cfu compared to the alginate formulation using 10% wheat flour. Yeast extract amendment further increased the number of cfu by about 38 and 32% for FN and FA, respectively, compared to sorghum amendment. In the first field experiment (2003/04), a screening for the suitable dose of ?Pesta? granules per planting hole to control Striga was conducted together with the investigation of a seed coating treatment as an alternative delivery system of the biocontrol agents. The ?Pesta? technology showed a potential to be used as a delivery system to control S. hermonthica under field conditions. Both ?Pesta?-formulated Fusarium isolates were able to delay Striga emergence, reduce the total number of Striga shoots and induce disease symptoms on all growth stages of Striga plants, irrespective to the dose and method of application used. The highest control efficacy was achieved by applying FA at 1.5g, which reduced the total number of parasite shoots by 82 % and the number of healthy Striga shoots by 88% compared to the untreated control. As a consequence, sorghum biomass and sorghum 100-seed weight were increased by 86 and 110%, respectively. FN and the combination of the fungal isolates were slightly less efficient in controlling the parasites. 1.5 g ?Pesta? granules per planting hole was found to be the optimum dosage for Striga management since increasing the dosage did not result in a significant improvement of control. In the second season (2004/05), the efficacy of alginate formulations amended with 10% wheat flour applied at 1.5g/planting hole was evaluated in addition to the ?Pesta? formulation for controlling Striga under field conditions. Alginate granules were able to delay Striga incidence significantly and reduce the total number of Striga shoots by 64 ? 78 % compared to the control early in the season. In contrast to the first season, fungal isolates formulated in ?Pesta? granules had no pronounced effect on delaying Striga emergence, however, ?Pesta?-granulated Fusarium species were able to reduce the total number of Striga shoots by 42 ? 55 % compared to the control early in the season. By the end of the season, the effect of both formulations on the total number of Striga shoots became negligible, but they significantly increased disease incidence on Striga shoots compared to the untreated control. FA formulated in ?Pesta? or alginate pellets was especially effective in this regard, causing disease in 74 and 80% of the Striga plants and reducing the total number of healthy Striga shoots by 55 and 60% compared to the control, respectively. FA applied as ?Pesta? granules was the most effective treatment in reducing Striga biomass by 58 % compared to the control which was positively reflected in an increased sorghum grain yield (63%) and sorghum straw yield (73%) compared to the control. The reduction of the efficacy of the ?Pesta? formulation in controlling Striga in the second season compared to the first season can probably be attributed to three reasons. These include a) climatic conditions, which differed from the first season in higher temperatures coupled with lower rainfall and low relative humidity, b) sodicity problems in the fields which might have affected the proliferation of the fungi in the soil and c) an inhibitory effect of the metabolites of the applied insecticide Sevin (active ingredient Carbaryl (1-naphthyl N-methylcarbamate)) on the virulence of soil fungi. Furthermore, an outdoor pot experiment was conducted to study the efficacy of alginate formulations with different amendments (wheat flour, sorghum flour and yeast extract) in comparison to the ?Pesta? formulation and seed treatments on controlling Striga. In this experiment, FA formulated as ?Pesta? granules was the most effective treatment and successfully inhibited Striga emergence until the end of the season. This was reflected in a significantly increased sorghum plant height (by 80%) and sorghum shoot dry weight (400%) compared to the negative control. Fusarium species in alginate granules also delayed Striga emergence and reduced the total number of Striga throughout the growing season. The best efficacy was obtained by FA, which reduced the total number of Striga shoots by 71% (using 10% wheat flour) and 84% (6% sorghum flour or yeast extract) compared to the control. Likewise, it reduced the proportion of healthy Striga shoots by 71%, 88% and 84%, respectively, and Striga biomass by 50%, 81%, and 89%, respectively. Alginate formulations generally also significantly increased sorghum plant height by up to 80% and sorghum shoot dry weight by 200 to 400% compared to the control. It can therefore be summarized that of the investigated fungal isolates and granular formulations FA formulated in ?Pesta? granules showed the best efficacy in controlling Striga under field and controlled conditions. The ability of FA to produce trichothecene mycotoxins that could be a hazard to humans or animals was assessed from samples of the fungus growing on autoclaved wheat grains. Additionally, samples of harvested sorghum seeds from various plots inoculated with the biocontrol agents were investigated for the content of trichothecene mycotoxins. None of the following toxins were either produced by FA or translocated to harvested sorghum seeds under field conditions: nivalenol, fusarenon X, deoxynivalenol, 15-actetyldeoxynivalenol, scirpentriol, monoacetoxyscirpenol, diacetoxyscirpenol, T-2 triol, HT-2 toxin, T-2 toxin and neosolaniol. A simple seed coating treatment using fine ?Pesta? granules and gum Arabic as adhesive material also showed a potential to control Striga in the first season experiment. It was able to reduce the total number of Striga shoots by more than 55% compared to the control. Consequently, sorghum biomass was increased by 54 ? 67% and sorghum100-seed weight by 70%. The effects were comparable to that caused by chemical control using 2,4-D. To improve the efficacy of seed coating under field conditions for the second season experiments, the effect of five types of adhesive materials (2 types of cellulose, 2 types of organic polymers and a clay) on growth and sporulation of FA and FN were tested in solid and liquid media. Cellulose 1 and 2 were found to enhance radial growth of both isolates but did not increase sporulation in liquid media. Organic polymer 2 was found to retard both radial growth and sporulation of the two isolates. Organic polymer 1 and clay significantly enhanced the production of chlamydospores, especially by FA, compared to Richard?s solution alone or amended with the other tested materials. For this reason one of them was suggested to be used for seed coating in addition to Arabic gum. Sorghum seeds were coated by a private company (SUET Saat- und Erntetechnik GmbH, Eschwege, Germany), using air-dried fungal propagules fermented on 1.5 % (w/v) sorghum straw. Arabic gum was observed to give a better coverage of sorghum seeds and higher cfu per seed (4 x 104 [FA] and 19 x104 cfu [FN]) compared to the second adhesive material. Coating sorghum seeds with the biocontrol agents did generally not result in a significant reduction of Striga shoots in the field, but some of the seed-coating treatments were very efficient in inducing disease symptoms on Striga shoots. Especially FN coated with Arabic gum and FA applied to the seeds in fine ?Pesta? granules increased the proportion of diseased Striga shoots significantly compared to the control by 79%. In the pot experiment, the fungal isolates applied as a seed coating neither resulted in a significant reduction of Striga shoots. Nevertheless, FA applied to sorghum seeds using the adhesive material provided by SUET reduced the total number of Striga plants by 52% compared to the control at the end of the season. FN in the same treatment significantly increased the proportion of diseased Striga by 77% at the end of the season. The effect of seed coating on increasing sorghum plant height and dry matter was lower than that caused by the granular formulations and not statistically significant compared to the control. From the obtained results it can be concluded that both granular formulations applied to the planting holes and seed coating can be used as effective delivery systems for biocontrol fungi and can be adopted under field conditions to reduce Striga infestation. However, the granular formulations showed a higher efficacy in controlling Striga. Inoculum type and concentration as well as nutritional amendments to the formulations should be further optimized in future investigations.Striga hermonthica ist eine parasitische Blütenpflanze aus der Familie der Orobanchaceae. Sie ist ein Wurzelparasit und befällt Sorghum, Mais, Hirse und ver- schiedene Gräser in den semiariden Tropen. Im Sudan ist Striga im Bewässerungs- und Regenfeldbau weit verbreitet und gilt als wichtigster biotischer Begrenzungsfaktor bei der Produktion von Sorghum, dem Hauptnahrungsmittel für die Mehrheit der sudanesischen Bevölkerung. Mehr als 500.000 Hektar des im Regenfeldbau kultivierten Ackerlands sind stark mit Striga befallen, was in der Regel Ertragsverluste von 70 bis 100% zur Folge hat. Mittlerweile ist deutlich geworden, dass es für das Striga-Problem in Afrika keine einfache, schnelle und billige Lösung gibt. Biologische Kontrolle gilt als ein potentiell kostengünstiges und umwelt- freundliches Instrument, um Unkrautpopulationen in Ackerkulturen, Wäldern oder Weideland, wo niedrige Gewinnspannen einen hohen Herbizideinsatz unmöglich machen, zu regulieren. Die biologische Kontrolle von S. hermonthica mit Mikro- organismen (v.a. phytopathogenen Pilzen) erwies sich in bisherigen Untersuchungen als effektiv. Bis jetzt wird allerdings kein derartiges Verfahren in der Praxis genutzt, was unter anderem auf den Mangel an geeigneten Formulierungs- und Applikationssystemen zurückzuführen ist. Die Pathogenität zweier Fusarium-Isolate aus dem Sudan (Fusarium nygamai (FN) und Fusarium ?Abuharaz? (FA)-Isolat) gegenüber Striga wurde mit Hilfe infizierter Sorghumkörner oder einer Sporensuspension als Inokulum untersucht. Mit diesen Formulierungen wurde Striga sehr effektiv unter kontrollierten und Freilandbedingungen bekämpft, allerdings wurden für eine ausreichende Kontrolle sehr große Mengen an Inokulum (etwa 800 kg ha-1 infizierte Sorghumkörner) benötigt. Dies würde für einen großflächigen Einsatz beispielsweise hinsichtlich Sterilität und Transport sehr problematisch werden, daher ist die Entwicklung einer angepassten Formulierungstechnologie unumgänglich. Granuläre Formulierungen wie ?Pesta? oder Alginat-Pellets können für die biologische Unkrautbekämpfung geeignet sein. ?Pesta?-Granulate werden durch Einkapseln der biologischen Gegenspieler in eine Glutenmatrix hergestellt. Für die Produktion von Alginat-Pellets wird das Inokulum in eine Natriumalginatlösung eingebracht, die in Calciumchlorid oder -gluconat ein- getropft wird. Die Pellets enstehen dann durch ionotrophe Gelbildung. Die Hauptziele der vorliegenden Arbeit waren: a) die Effizienz der zwei Fusarium-Isolate formuliert als ?Pesta?-Granulate oder Alginat-Pellets hinsichtlich ihrer Wirkung auf Striga unter Feldbedingungen zu untersuchen; (b) den Effekt auf den Sorghumertrag zu evaluieren; (c) die optimale Dosis des formulierten Materials zu bestimmen; (d) die Persistenz der formulierten pilzlichen Isolate im Boden zu beobachten; und (e) die Effizienz von Saatgutbehandlungen als alternative Applikationsform zu untersuchen. Hinsichtlich der Umweltverträglichkeit der potentiellen Produkte wurde das erst kürzlich isolierte F. ?Abuharaz? auf seine Eigenschaft getestet, einige der wichtigsten Mykotoxine zu produzieren. Geerntete Sorghumkörner aus den Fusarium-Behandlungen im Feldversuch wurden ebenfalls hinsichtlich ihres Mykotoxingehaltes überprüft. Eine Voraussetzung für die Formulierung von biologischen Gegenspielern ist die Entwicklung einer preiswerten Methode zur Produktion von Inokulum, die es ermöglicht, ausreichend pilzliche Biomasse mit lebensfähigen, virulenten Vermehrungseinheiten zu gewinnen. Chlamydosporen sind die Überdauerungs-formen vieler Fusarium-Arten und werden als ideale Sporenform für die Herstellung von Festformulierungen betrachtet. Daher war eines der speziellen Ziele dieser Arbeit, ein Medium zu finden, das die beiden untersuchten Fusarium-Isolate zur Chlamydosporenproduktion anregt. Verschiedene Flüssigmedien wurden getestet, von denen SNB (Special Nutrient-poor Broth) plus Hefe bei beiden Isolaten die besten Ergebnisse erzielte (105 Chlamydosporen ml-1). Diese Anzahl war jedoch noch zu gering, um für die Weiterverarbeitung in einer Formulierung in Frage zu kommen. Die höchste Anzahl an Mikrokonidien nach 5 Tagen Inkubationszeit (108 ml-1) wurde in Richard?s Medium gebildet, daher wurde dieses während der ge- samten Arbeit als Wachstumsmedium für die Pilze verwendet. FN und FA wurden erfolgreich sowohl in ?Pesta?-Granulate als auch in Alginat-Pellets (angereichert mit 10% Weizenmehl oder 6% Sorghummehl oder Hefeextrakt) formuliert. Alginat-Pellets zeigten durchweg eine höhere Anzahl von ?colony forming units? (cfu) pro g formuliertem Material verglichen mit den ?Pesta?-Granulaten. Die Alginat-Formulierungen mit 6% Sorghummehl oder Hefeextrakt hatten signifikant höhere cfu verglichen mit den Pellets mit Zusatz von 10% Weizenmehl. Der Zusatz von Hefeextrakt erhöhte die Anzahl der cfu pro g nochmals um 38% (FN) und 32% (FA) gegenüber dem Zusatz von Sorghummehl. Im ersten Feldversuch (2003/2004) wurde ein Screening für eine effektive Dosis der ?Pesta?-Granulate durchgeführt, zusätzlich wurde die Saatgutbehandlung als alter- native Applikationsmethode der biologischen Gegenspieler getestet. Die ?Pesta?-Formulierung erwies sich als geeignet für die Applikation der Fusarium-Isolate. In dieser Formulierung verzögerten beide Isolate den Auflauf der Striga-Pflanzen, ver- ringerten die Gesamtanzahl der Sprosse und induzierten Krankheitssymptome an allen Stadien der Parasiten, unabhängig von der eingesetzten Dosis. Die höchste Kontrolleffizienz wurde nach der Applikation von FA mit 1.5 g pro Pflanzloch erzielt: die Gesamtanzahl der Striga-Sprosse gegenüber der unbehandelten Kontrolle wurde um 82% und die Anzahl der gesunden Striga-Sprosse um 88 % reduziert. Dies hatte eine Erhöhung der Sorghum-Biomasse und des 100-Korn-Gewichts von Sorghum um 86% bzw. 110% zur Folge. FN und die Kombination beider Fusarium-Isolate kontrollierten das parasitische Unkraut mit einer leicht geringeren Wirksamkeit. 1.5 g ?Pesta?- Granulat pro Pflanzloch erwies sich als ausreichende Dosis für das Striga- Management, da durch Erhöhung der Dosierung keine weitere Wirksamkeits- steigerung erzielt werden konnte. In der zweiten Feldversuchsperiode (2004/2005) wurde zusätzlich zu den ?Pesta?-Granulaten die Effizienz der Alginatformulierungen mit 10 % Weizenmehl in einer Dosis von 1.5 g pro Pflanzloch untersucht. Die Alginat-Pellets konnten den Striga-Befall signifikant verzögern und reduzierten die Anzahl der Sprosse anfangs um 64-78% gegenüber der unbehandelten Kontrolle. Im Gegensatz zum ersten Versuch verzögerten die pilzlichen Isolate in ?Pesta?-Granulaten das Auflaufen von Striga nicht signifikant, allerdings konnten sie die Anzahl der Sprosse anfangs um 42-55% gegenüber der Kontrolle reduzieren. Am Ende der Vegetationsperiode wurde der Effekt beider Formulierungen auf die Anzahl der Striga-Sprosse vernachlässigbar gering, aber sie erhöhten signifikant den Anteil erkrankter Sprosse im Vergleich zur unbehandelten Kontrolle. FA in Alginat-Pellets oder als ?Pesta? war in dieser Hinsicht besonders effektiv, das Isolat verursachte bei 74 und 80% der Sprosse Krankheits- symptome und konnte so die Anzahl gesunder Sprosse um 55 und 60% gegenüber der Kontrolle reduzieren. FA in ?Pesta? war auch die effektivste Behandlung hinsichtlich der Reduktion der Striga-Biomasse (58% im Vergleich zur Kontrolle), was sich positiv in einem erhöhten Korn- (63%) und Strohertrag (73%) bei der Wirts- pflanze auswirkte. Die Unterschiede in der Kontrolleffizienz der ?Pesta?-Formulierung zwischen den bei- den Versuchsjahren sind möglicherweise auf drei Gründe zurückzuführen. Zum einen unterschieden sich die Klimabedingungen zwischen beiden Jahren, im zweiten Versuchsjahr herrschten deutlich höhere Temperaturen in Verbindung mit weniger Niederschlägen und einer geringeren relativen Luftfeuchtigkeit. Zum anderen traten im Feld im zweiten Versuchsjahr größere Probleme mit hoher Natriumsättigung des Bodens auf, was die Vermehrung der Pilze im Boden beeinträchtigt haben könnte. Zum dritten könnten sich die Abbauprodukte des applizierten Insektizids Sevin (Carbaryl (1-naphthyl N-methylcarbamat)) negativ auf die Virulenz der Isolate aus- gewirkt haben. Zusätzlich wurde ein Freiland-Topfversuch durchgeführt, um die Effizienz der Alginat-Formulierungen mit verschiedenen Zusätzen (Weizenmehl, Sorghummehl und Hefe- extrakt) hinsichtlich der Striga-Kontrolle im Vergleich mit der ?Pesta?-Formulierung und den Saatgutbehandlungen zu untersuchen. In diesem Versuch war die Applikation von FA in ?Pesta?-Granulaten die effektivste Behandlung und verhinderte das Auflaufen von Striga bis zum Ende der Vegetationsperiode. Dies spiegelte sich in signifikant höheren Wirtspflanzen (um 80%) und erhöhtem Sorghum-Spross- trockengewicht (400%) gegenüber der negativen Kontrolle wider.     Investigations of the pre-treatment and the conversion of energy crops into biogas and bioethanol

    Da die fossilen Ressourcen endlich sind, ist eine Option für die Zukunft den Energiebedarf verstärkt aus erneuerbaren Energiequellen zu decken. Eine Möglichkeit ist die Biomassenutzung, die durch eine Vielzahl von Kombinationen aus unterschiedlichen Biomassearten, Nutzungspfaden und Konversionstechniken flexibel an die natürlichen lokalen bzw. regionalen Gegebenheiten sowie die anthropogenen Bedürfnisse angepasst werden kann. Um die begrenzten landwirtschaftlichen Flächen möglichst effizient zur Bioenergieträgerbereitstellung nutzen zu können, werden aktuelle und belastbare Daten zu den spezifischen Energieerträgen und den Hektarenergieerträgen von Energiepflanzen benötigt. Ziel der vorliegenden Arbeit war es, diese Daten für die Bereiche Biogas und Bioethanol bereitzustellen. Dazu wurden anhand von Untersuchungen in Laborfermentern im Batch-Betrieb spezifische Biogas- bzw. Bioethanolerträge ermittelt. Zusätzlich stand die Erprobung von verschiedenen Aufbereitungsverfahren für Energiepflanzen und deren Einfluss auf die Biogasertragshöhe und auf den Verlauf der Methanbildung im Fokus dieser Arbeit. Eine Energie- und Ökobilanz für Silomais und reife Triticale stellt die Konversionspfade Biogas- und Bioethanolgewinnung vergleichend gegenüber. Dabei wurde auch das Steam-Explosion-Verfahren zur Aufbereitung von Biomasse berücksichtigt. Aufbereitung von Energiepflanzen Die Aufbereitung von Biomasse mit dem Steam-Explosion-Verfahren beschleunigt die Methanbildung und steigert sie zum Teil. Die Effekte sind, abhängig von der Art der Biomasse und dem Reifestadium, unterschiedlich stark. Weitere Aufbereitungsverfahren wie Mikrowellenbehandlung und Kochen zeigten meist keine signifikante Änderung oder teilweise negative Wirkung. Eine Variation der Versuchsparameter könnte aber ggf. interessant sein. Den positiven Wirkungen des Steam-Explosion-Verfahrens stehen aber auch Argumente wie die zusätzlichen Investitionskosten und der Verdünnungseffekt durch die Wasserzugabe bzw. die Erhöhung des Massenstroms entgegen. Der zusätzliche Energiebedarf, der hauptsächlich in thermischer Energie besteht, kann aus der Abwärme des BHKW gedeckt werden. Forschungsbedarf besteht weiterhin beim Screening und der Produktion technischer Enzyme zum effizienten Voraufschluss lignozellulosehaltiger Rohstoffe sowie bei der Kombination biologischer (enzymatisch), chemischer, thermischer oder mechanischer Aufschlussverfahren unter Berücksichtigung der Energieeffizienz. Methanerträge von Energiepflanzen und Schlempen Anhand der Biogasuntersuchungen eines breiten Spektrums an Maissorten konnte festgestellt werden, dass die spezifischen Methanerträge je nach Sorte unterschiedlich stark über die Erntezeitpunkte variierten, wobei die Sorten mit niedrigerer Reifezahl höhere spezifische Methanerträge erreichten. Der dominierende Faktor für den Energiehektarertrag war aber der Trockenmasseertrag und nicht der spezifische Methanertrag. Allgemein empfehlenswert sind standortgerechte Sorten mit hohem Trockenmasseertrag bei gleichzeitig guter Silierfähigkeit zum optimalen Erntezeitpunkt. Die Zwischenfrüchte trugen nur zum Teil zur deutlichen Erhöhung der Methanhektarerträge bei. Aus Gründen des Bodenschutzes ist der Anbau aber zu empfehlen. Die Stickstoffdüngung beeinflusst die Trockenmasseerträge und damit die Energiehektarerträge meist positiv. Mit Mais konnten deutlich höhere Energiehektarerträge als mit Rutenhirse erzielt werden. Durch die Nutzung der Schlempen aus der Ethanolproduktion aus Maisganzpflanzen oder aus Triticalekorn sowie durch die Nutzung der Nebenprodukte wie Stroh in der Biogasgewinnung kann der Energieoutput pro Hektar mindestens verdoppelt werden verglichen mit der ?reinen? Ethanolproduktion. Weitere Optimierungsmöglichkeiten für die Biogasgewinnung unter Praxisbedingungen liegen im substratangepassten Aufbau der Fermentersysteme sowie im Einsatz mehrphasiger Verfahren sowie der Entwicklung schneller Analyseverfahren zur besseren Prozesssteuerung. Korrelation Inhaltsstoffe / gemessene Methanerträge Die anhand der über NIRS bestimmten Inhaltsstoffe neutrale Detergentien-Faser (NDF), Stärke (XS), Zucker (XZ) und Rohprotein (XP) sowie der substrattypischen Faktoren errechneten spezifischen Methanerträge für Maisproben zu den vier Erntezeitpunkten lagen sehr nah an den tatsächlich gemessenen Methanerträgen. Eine Korrelation zwischen den gemessenen und errechneten Werten lag aber nicht vor. Ob eine andere Inhaltsstoffanalysetechnik und die Bestimmung weiterer Einzelbestandteile die Biogasertragstests zur Potenzialabschätzung gerade von neuen Sorten ersetzen können, sollte Gegenstand weiterer Untersuchungen sein. Bioethanolausbeuten Die Energieausbeuten über den Konversionspfad Bioethanol liegen ohne die Nutzung der Nebenprodukte Schlempe oder Stroh deutlich unter denen des Konversionspfads Biogas, da nur Stoffe, die verzuckert werden können, in Ethanol umgesetzt werden können. Der Vorteil der Ethanolproduktion liegt in dem flüssigen Kraftstoff, der gewonnen wird. Durch die Kombination mit einer Biogasanlage können auch die Nebenprodukte energetisch genutzt werden und liefern einen gasförmigen Energieträger. Im Bioethanolbereich liegen sicher noch hohe bioverfahrenstechnische Optimierungspotenziale z.B. die Züchtung geeigneter Mikroorganismen zum Abbau lignozellulosehaltiger Rohstoffe sowie von C5-Zucker-Verwertern. Des Weiteren sind Prozessoptimierungen bei Wasser- und Energieeinsatz anzustreben. Energie- und Ökobilanzierung Sowohl Biogas als auch Bioethanol, letzteres bei optimaler Kombination mit Biogas, sind nach den hier untersuchten Szenarien unter den erläuterten Rahmenbedingungen dazu geeignet, den Einsatz nicht regenerativer Energieträger und damit Emissionen zu senken. Eine Aufgabe für die Zukunft wird es sein, differenzierte standortgerechte Nutzungskonzepte zu entwickeln auf der Basis einer Entscheidung, in welchem Maß eine (Flüssig-)Kraftstofferzeugung oder eine stationäre Bereitstellung thermischer und elektrischer Energie aus Biomasse zu bevorzugen sind. Ziel muss es sein, durch eine sinnvolle Kombination aus Biomasse, Nutzungspfad und Konversionstechnik, in Abhängigkeit von den lokalen und regionalen natürlichen Gegebenheiten sowie den anthropogenen Anforderungen, die land- und forstwirtschaftliche Fläche (als dem limitierten Faktor) höchst effizient zur Bioenergieträgerbereitstellung zu nutzen.Due to finite fossil resources, one opportunity for the future is to increase the supply of energy out of renewable energy sources. One of many opportunities is the use of biomass, which offers plenty combinations of different kinds of biomass, paths of utilization and conversion techniques for a flexible adaptation to natural local and regional frameworks as well as the anthropogenic needs. For an efficient utilization of the limited arable land for the supply of bioenergy, there is a need of up-to-date and proof data about specific energy yields and yields per hectare. The aim of this investigation was to determine these data for the biogas and bioethanol sectors. Batch-tests were carried out in laboratory scaled digesters to investigate specific biogas and bioethanol yields. Additionally the testing of different techniques of pre-treatment for energy crops and their effects on the biogas yield and the progression of the formation of methane were focused. The conversion of maize silage and full ripe triticale into biogas and bioethanol was compared by an energy and environmental balance. The steam explosion technique was included. Pre-treatment The steam explosion pre-treatment of biomass increases the speed of formation of methane and partly increases the methane yields. The effects differ depending on the kind of biomass and the stage of ripening. Other techniques of pre-treatment like microwaving and cooking did not show significant or partly negative effects. A variation of parameters in the trial setup might be interesting. Besides the positive effects of the steam explosion technique there are some arguments like the additional costs of investment, the diminished concentration of nutrients respectively the increase of material flow against it. The additional energy consumption, mostly thermal energy, can be supplied from waste heat out of the combined heat and power plant (CHP). The screening and the production of technical enzymes for the efficient pre-degradation of raw materials containing high amounts of lignocellulose should be the subject of research and development in the future. The combination of biological (enzymatic), chemical, thermal and mechanical pre-treatment techniques need to be investigated with the focus on energy efficiency. Methane yields of energy crops and stillage A broad number of biogas tests had been carried out on various maize cultivars. The specific methane yields of the maize cultivars varied over the harvesting date differently. The cultivars with a low ripening number reached higher specific methane yields. The dominant factor for the energy yield per hectare was the dry matter yield, not the specific methane yield. In general it is recommended to use well adapted cultivars with high dry matter yields and a good ensilaging behaviour. The catch crops increased the methane yields per hectare just partly. But for reasons of soil conservation the cultivation is recommended. The nitrogen fertilizer had mostly a positive effect on the dry matter yields and the energy yields per hectare, respectively. Maize gained higher energy yields per hectare than switch grass. The utilization of stillage out of whole maize plants or triticale´s grain from the ethanol production as well as the utilization of by-products like straw in the biogas production could double the energy output per hectare compared to the simple ethanol production. Further options for the optimization of the biogas production under conditions of practice are digester systems well-adapted on the substrate, the use of multi step systems and the development of analytic methods in order to gain effective process control. Correlation between chemical components and measured methane yields The specific methane yields calculated out of the neutral detergents fibre, starch, sugar, raw proteins and its substrate-specific factors were very close to the experimentally determined yields of the maize cultivars for the four harvesting times. But the measured and calculated values showed no correlation. Whether the biogas tests can be replaced, by other methods or techniques of analysis of the components and the determination of additional components for the estimation of the potential of new cultivars, should be subject of further investigations. Bioethanol yields The energy yields on the conversion pathway bioethanol without using the by-products are lower than the yields via conversion into biogas, because the ethanol fermentation is limited on material that can be converted into sugar first. The advantage of the ethanol production is a fluid fuel as result of the process. Combining the ethanol production with a biogas plant, the by-products also can be used energetically and a gaseous energy carrier can be produced. There are high potentials for the bioprocess engineering, for instance in breeding of microorganisms for the degradation of lignocellulosic biomass or of C5-sugar. Furthermore a process optimization of water and energy input is recommended. Energy and environmental balance Biogas as well as bioethanol (combined with biogas) is able to reduce the consumption of non-renewable energy carrier and its emission under the investigated scenarios and the scoop set. A future task will be the development of differentiated and well-adapted concepts on the basis of a decision between (liquid) fuels or stationary supply of thermal and electrical energy out of biomass. The aim is an efficient use of the limited areas of arable land and forests for the supply with bioenergy carriers by a useful combination of biomass, paths of utilization and conversion technique depending on natural local and regional conditions as well as the anthropogenic needs

    Identification and functional characterization of succinic semialdehyde dehydrogenase from parasitic and nonparasitic arthropods

    Ziel dieser Arbeit war es, die in Insektenmodellen weitgehend und in parasitischen Insekten und Akariden vollständig unerforschte SSADH molekularbiologisch und enzymologisch zu charakterisieren. Als Organismen der Wahl dienten das Bakterium Escherichia coli (Ec) und die Maus Mus musculus zu Referenzzwecken und für biophysikalische Untersuchungen des Enzyms, sowie Drosophila melanogaster (Dm) (Fruchtfliege), Lucilia cuprina (Lc) (Schafs-Schmeissfilege), Ctenocephalides felis (Cf) (Katzenfloh), und Rhipicephalus microplus (Rm) (Zecke) als Vertreter verschiedener Organismenklassen. Für Dm lag zunächst durch ?Expressed Sequence Tag?-Studien sowie der Entschlüsselung des Genoms ein Einzelgenkandidat vor, für den Homologiebetrachtungen nahe legten, dass es sich um das Fruchtfliegen-Genortholog für SSADH (DmSSADH) handeln könnte. Im ersten Teil dieser Arbeit wurde dieser Genkandidat kloniert, und in Ec exprimiert. Zur vergleichenden Charakterisierung der enzymatischen Eigenschaften des SSADH-Expressionsprodukts wurde ein Genkandidat für eine Acetaldehyd-Dehydrogenase (DmALDH) aus Dm kloniert und in Ec exprimiert. Beide Produkte zeigten erwartete enzymatischen Eigenschaften: NAD+-abhängige Succinsemialdehyd-oxidierende (DmSSADH) und NAD+-abhängige Acetaldehyd-oxidierende Aktivität (DmALDH). Ein Vergleich der Substrateigenschaften von 30 Aldehyden und z.T. enzymkinetischen Parameter belegte die ausgeprägte Spezifität von DmSSADH für Succinsemialdehyd, während DmALDH als unspezifische Aldehyddehydrogenase eingeordnet wurde. Punktmutagenesestudien zeigten, dass zwei für die SSADH-Aktivität essentielle Aminosäuren Glutamat 277 und Cystein 311 sind. Im zweiten Teil der Arbeit erfolgte die Genidentifikation, Volllängen-Genklonierung, sowie die funktionelle Expression jeweils eines Genvertreters von SSADH aus Lc und Cf. Enzymkinetische Studien und Aktivitätsuntersuchungen der E. coli-exprimierten L. cuprina- (LcSSADH) und C. felis- (CfSSADH) Enzyme zeigten, dass beide vor allem NAD+-abhängige SSADHs sind. Analysen der LcSSADH und CfSSADH-Gene mittels PCR-und Southern-Blots zeigten, dass das SSADH-Gen in beiden Organismen nicht als Einzelkopie vorliegt, und dass die Exon-Intron-Struktur von Dm nicht konserviert ist. Die Funktionen der aufgedeckten multiplen Genkopien und die Gründe für die variablen Exon-Intron-Strukturen in den SSADH-Genen der hier betrachteten Spezies bleiben unklar. In Immunblot-Experimenten konnte schließlich gezeigt werden, dass SSADH in Imagines von Dm, Lc und Cf auch auf Proteinebene exprimiert ist. Der dritte Teil der Arbeit hatte die Genidentifizierung, sowie die biochemische Charakterisierung der SSADH aus der Zecke Rm zum Thema. Southern-Blot-Analysen zeigten, dass das SSADH-Gen im Rm-Genom wahrscheinlich in 2-3 Genkopien vorhanden ist. Für einen Vergleich der Zecken-SSADH mit dem Genprodukt eines Mammalier-Orthologen, wurde das SSADH-Gen aus der Maus isoliert und funktionell in Ec exprimiert. Der Vergleich der beiden Enzym ergab, dass beide potente NAD+-abhängige Succinsemialdehyd-oxidierende Aktivität besitzen und sehr ähnliche enzymkinetische Parameter aufweisen. Eine genauere vergleichende Betrachtung der Aktivität gegenüber verschiedenen Aldehydsubstraten zeigte deutliche Unterschiede zwischen den Enzymen beider Spezies. Generell scheint Maus-SSADH spezifischer für Succinsemialdehyd zu sein als Rm SSADH. Weiterhin ist das Mammalierenzym gegenüber den in dieser Studie identifizierten Inhibitoren p-Tolualdehyd und p-Methoxybenzaldehyd ebenso wie gegenüber Substratinhibition durch einen Überschuss Succinsemialdehyd weit weniger empfindlich als das Zeckenenzym. Es bleibt zu untersuchen, ob diese Unterschiede für die Auffindung potenter und spezifischer Inhibitoren nutzbar sind. Ebenso bleibt die Frage zu beantworten, ob SSADH, bzw. der GABA-Abbau allgemein eine valide akarizide Targetstruktur darstellt. Im vierten Abschnitt der Arbeit wurden drei im Rahmen dieser Arbeit hergestellte SSADH-Enzympräparate in Sättigungstransfer-Differenz-Kernresonanzspektroskopie-(STD-NMR-)Experimente eingesetzt, um Fragen zur Aldehydsubstrat- und Cosubstratbindung an SSADH zu beantworten. Es konnte eindeutig gezeigt werden, dass bei Ec und Dm SSADH das freie Aldehyd dem in wässriger Lösung für Succinsemialdehyd in gleicher Konzentration vorliegende geminale Diol (?gem-Diol?) als Substrat bevorzugt. Die STD-NMR-spektroskopische Untersuchung der Succinsemialdehyd-Interaktion mit der Dm SSADH C311A-Mutante zeigte die Bindung sowohl der Aldehyd- als auch der gem-Diol-Form. Dies deutet darauf hin, dass C311 für die Aldehyd-Selektivität des Enzyms entscheidend ist. Das STD-NMR-Epitopmapping der NAD+/NADP+-Bindung an E. coli-SSADH und an Dm SSADH legte in beiden Fällen die dominanten Protein-Interaktionen der Adenin- und Nikotinamidringe nahe, während die Ribose-Reste vermutlich weit weniger intensiv mit dem Enzym verbunden sind.The objective of the PhD project was the molecular and enzymological characterization of SSADH in parasitic and nonparasitic insects and acarids, organism groups where this enzyme was virtually unexplored. In this project, the list of investigated non-arthropod organisms included the bacterium Escherichia coli (Ec) and the mammal Mus musculus (mouse). Their SSADH served as reference enzymes or were used for biophysical experiments. Amongst the arthropods, Drosophila melanogaster (Dm) (fruit fly) was chosen as nonparasitic model insect. Lucilia cuprina (Lc) (sheep blowfly) and Ctenocephalides felis (Cf) (cat flea) were included as important parasitic insects, as was the acarid Rhipicephalus microplus (Rm) as the economically most important tick species. At the start of this thesis, expressed sequence tag studies and whole genome sequencing on Dm suggested the existence of a single copy gene candidate that, based on amino acid sequence homology, was considered to be a candidate for a SSADH ortholog in this species (DmSSADH). The Dm SSADH gene candidate was cloned and expressed in Ec as a soluble protein. To compare the enzymological properties of DmSSADH, another so far uncharacterized Dm gene candidate for an acetaldehyde dehydrogenase (DmALDH) was cloned and expressed in Ec as a soluble protein. Both expression products showed the expected enzymological properties: a NAD+-dependent ssa-oxidizing activity for DmSSADH and a NAD+-dependent acetaldehyde-oxidizing activity for Dm-ALDH. Site-directed mutageneses on DmSSADH performed in this study demonstrated that two residues essential for catalysis are glutamate 277 and cysteine 311. The second part of the thesis encompassed the gene identification, full length gene cloning of SSADH in Lc (LcSSADH) and Cf (CfSSADH), as well as functional expression of one gene version, respectively. Substrate/cosubstrate specificity determinations combined with enzyme kinetics studies showed that both enzymes are predominantly NAD+-dependent SSADHs. Bioinformatics analyses detected N-terminal mitochondrial import sequences in both Lc and Cf SSADH suggesting that these enzymes are localized in vivo in the mitochondrial matrix. The investigation of the genomic structure of the LcSSADH and CfSSADH genes revealed significant differences to the previously known gene organizations: firstly, different to the single copy SSADH gene situation in Ec, Dm, Mus musculus and Homo sapiens, Lc appears to possess 2-3 SSADH genes, while in Cf up to 8 gene copies may be present. Secondly, compared to Dm (2 exons, 1 intron), the exon/intron structure of the SSADH genes in Lc and Cf is not conserved: the one SSADH genomic gene version investigated in detail in Lc contained 3 exons and 2 introns, while the genomic gene version of Cf analysed in this study was devoid of any introns. The central topic of the third part of this thesis was the gene identification and biochemical characterization of SSADH from the tick Rm. By a combination of different PCR methods a tick gene orthologous to insect and mammalian SSADH (RmSSADH) could be identified and isolated. The results of Southern blot analyses of Rm DNA are incompatible with a single copy situation and suggest the presence of 2-3 RmSSADH genes. To compare RmSSADH with a mammalian SSADH, the respective gene was isolated from mouse. Both the tick and the mouse SSADH genes were then expressed as soluble functional proteins in Ec. The initial comparison showed that both proteins are potent NAD+-dependent ssa-oxidizing enzymes with very similar enzyme kinetics. A more detailed comparison of both enzymes suggests that in general, the mouse enzyme appears to be more specific for succinic semialdehyde than the tick enzyme. In the fourth section of the experimental part of this thesis, saturation transfer difference nuclear magnetic resonance spectroscopy (STD-NMR) experiments were performed on three of the above SSADH preparations, to answer questions on the aldehyde substrate and cosubstrate binding to these enzymes. Importantly, the long-standing question whether the free aldehyde of succinic semialdehyde or its hydrated gem-diol form (present in aqueous solution in equimolar amounts) is the binding substrates was answered conclusively in favour of the aldehyde form for both the Ec and the Dm enzyme. Most remarkably, STD-NMR experimental investigation of the ssa interaction with the Dm SSADH cysteine311alanine mutant enzyme demonstrated binding of both the aldehyde and the gem-diol form. This experiment strongly suggests that cysteine311 is acting as an aldehyde versus gem-diol selectivity filter in the active site of the enzyme. Furthermore, STD-NMR epitope mapping of the NAD+/NADP+ binding to Ec SSADH and Dm SSADH was performed. In both cases, the data suggested that the dominant protein-ligand interactions are via the adenine and the nicotinamide ring systems, while the ribose moieties interact much less intensely with the enzymes

    Structural change requirements in the Bulgarian dairy sector aiming at higher competitiveness within the EU

    During the burdensome years of transition the agriculture in Bulgaria plays the role of a social buffer and a sector providing some, although insufficient, income and employment. Although employment in agriculture is a source of income, self consumption of its products can save income that could be spend on something else. The differences between market and self-sufficiency oriented farmers diminish due to income instability, that consequently contribute to agricultural decommercialization. A major characteristic of small-scale subsistent farming is the diversification of production activities that usually lead to diseconomy of scale effects. At the same time small-scale subsistent farms use labour intensive systems of production as a substitution for the scarcity of capital and machinery. Subsistence farming uses resources which could be used elsewhere in market-oriented farming and other sectors and its existence may cause a loss of overall production efficiency. Notwithstanding this loss of efficiency at the aggregate level, subsistence farmers may be efficient with regard to their own utility functions. Consequently, from a conventional economics point of view, small-scale farmers are unlikely to react to government policies in a normal, "rational" way. However, when they dominate the production of some products, predictions based on ?normal? economic models may be unreliable. The scope of the study is to cover the agriculture holdings with dairy cows according to the national statistic and moreover to argue that not all of them can be defined as dairy farms. The general hypothesis of this thesis states that the current typical dairy farm can double its size and increase significantly its income while reducing the risk for the household. On the contrary if it is growing more than a double that would have the opposite effect due to overestimated management capacity and unacceptable size of liabilities. The method used in this thesis is based on the concept of a typical dairy farm through bottomup approach. A typical dairy farm represents a significant number of dairy farms in a region in terms of size, forage and crops grown, livestock systems, labour organization and production technology used, and show an average management / performance ratio. The typical farm is ?built? and ?validated? based on panels (farmers, advisors? knowledge and local experts) and farm accounting statistics. The simulations in this thesis proved that with the currently existing support programs a successful farm restructuring is viable in a short period of time if the farmer possesses the necessary skills, knowledge and information to adopt a strategy to successfully face the changing market conditions. While the suggested structural changes could be successfully implemented in order to provide a significant improvement of the management, the time span available for them is very ?narrow? with respect to the financial support provided by the programs available. The general assumption of the government policy was that the ?Producer Union? (PU) should play a leading role in the process of structural reforms in agriculture. Unfortunately that assumption didn?t justify itself, consequently the provision of high qualified management services (as a major benefit from the membership in the PU) to the farmers is not utilised by them.Während der beschwerlichen Jahre der politischen Wende kam der Landwirtschaft in Bulgarien die Rolle eines sozialen Puffers zu, welcher der Bevölkerung, allerdings in unzureichendem Maße, Einkommen und Arbeit bot. Die Beschäftigung in der Landwirtschaft stellt eine Einkommensquelle dar, gleichwohl kann durch den Eigenkonsum der Erzeugnisse Einkommen gespart werden, welches für andere Zwecke verwendet werden kann. Instabilitäten im Einkommen führen zu einer Verringerung der Unterschiede zwischen marktorientierten und selbstversorgungsorientierten Bauern, was zur Dekommerzalisierung der Landwirtschaft beiträgt. Ein Hauptcharakteristikum kleiner Subsidenzbetriebe ist die diversifizierte Produktion, die üblicherweise der Erzielung positiver Skaleneffekte gegenüber steht. Gleichzeitig herrscht in kleinen Subsistentbetrieben, als Substitut für die knappen Ressourcen Kapital und Arbeitsmaschinen, ein arbeitsintensives Produktionssystem vor. Die Verpflegung der Agrarwirtschaft verwendet Ressourcen, die anderswo in marktorientierter Agrarwirtschaft und anderen Sektoren Gebrauch finden. Ihre Existenz kann zur Verringerung der gesamten Produktionseffizienz führen. Trotz der Effizienzverringerung auf dem aggregierten Niveau, können Subsistenzlandwirten bzgl. ihrer Nutzfunktionen effizienter werden. Als Konsequenz, vom konventionellen ökonomischen Standpunkt aus, können mittelständische Landwirten auf Regierungspolitik nicht ?rational? reagieren. Trotzdem, falls sie die Produktion von einigen Produkten dominieren, können Prognosen, die auf standarden ökonomischen Modellen basieren, nicht verlässlich sein. Die Hauptthese der Arbeit besagt, dass der typische derzeitige Milchviehbetrieb in der Größe verdoppelt und das daraus erzielbare Einkommen signifikant erhöht werden kann, bei gleichzeitig verringertem Risiko für den jeweiligen Haushalt. Würde der Betrieb dagegen um mehr als das Doppelte wachsen, ergäbe sich ein gegenteiliger Effekt aufgrund überschätzter Managementkapazitäten und einer inakzeptablen Verschuldungshöhe. Die Methodik der Arbeit basiert auf dem Konzept eines typischen Milchviehbetriebs. Ein typischer Milchviehbetrieb repräsentiert eine signifikante Anzahl an Milchviehbetrieben einer Region in Hinsicht auf die Größe, die Struktur von Futterbau und Feldfrüchten, die Haltungssysteme in der Tierproduktion, die Arbeitsorganisation und die verwendete Technologie und weist ein durchschnittliches Managment / Performance Verhältnis auf. Der typische Betrieb wird ?konstruiert? und ?validiert? basierend auf Panels (Landwirte, Expertise von Beratern und lokalen Experten) und landwirtschaftlichen Rechnungslegungsstatistiken. Zusammenfassend haben die Simulationen in dieser Arbeit gezeigt, dass unter den derzeit existierenden Förderprogrammen eine erfolgreiche Restrukturierung der Betriebe in einem kurzen Zeithorizont möglich ist, sofern die Landwirte die notwendigen Kenntnisse und Informationen besitzen, um eine Strategie zum erfolgreichen Bewältigen der sich ändernden Marktverhältnisse zu verfolgen. Während die vorgeschlagenen strukturellen Änderungen, um eine signifikante Verbesserung des Managements zu gewährleisten, erfolgreich implementiert werden könnten, ist die dafür verfügbare Zeitspanne hinsichtlich der finanziellen Unterstützung, die die Programme bieten, sehr begrenzt. Die generelle Annahme der Politik war, dass die Genossenschaften eine führende Rolle im Prozess der strukturellen Reform der Landwirtschaft spielen sollten.     Effect of low ethanol concentrations on the production and stability of Interferon gamma

    Alcohol is known to modulate the immune system in a complex manner. The effects of alcohol on immune responses vary with acute and chronic exposure as well as depending on the history of alcohol consumption and the blood level of alcohol. The presence or absence of alcohol can affect the cytokine cascade in complex ways. In the current study the immunmodulatory capability of an acute, moderate (1 ?) to high amount (3 ?) of alcohol was tested on isolated Peripheral Blood Mononuclear Cells production of several proinflammatory and antiinflammatory cytokines after incubation for 12 to 72 hours. The most affected cytokine in our model system of isolated human PBMC treated with two different ethanol concentrations was IFN-γ. Its concentration decreased in a highly significant manner in PHA- as well as in LPS-stimulated PBMC when treated with 66 mM ethanol and in a significant manner in PHA-activated PBMC when treated with 22 mM ethanol. The fact that ethanol negatively affects IFN-γ production is supported by several in vivo and in vitro studies by Wagner et al., 1992, Chen et al., 1993, Laso et al., 1997 Waltenbaugh et al., 1998, Starkenburg et al., 2001, Szabo et al., 2001, Dokur et al., 2003. The reduced IFN-γ level observed might be a key factor in explaining comprised immunity seen after chronic alcohol abuse, since together with IL-12, IFN-γ is crucial for the innate and adaptive immune response to viral and bacterial infection (Vicente-Gutierrez et al., 1991, Windle et al., 1993, Szabo 1997, Szabo et al., 1999). As seen in isolated human Peripheral Blood Mononuclear Cells IFN-γ production by IL-12 stimulated NK-92 cells is significantly reduced in the presence of ethanol. However, this decrease did not correlate with decreased phosphorylation and nuclear translocation of STAT4, a central regulator of IFN-γ gene expression. These results indicated that acute alcohol treatment in vitro did not affect intracellular pathways leading to IFN-γ gene expression. These findings paralleled results indicating that the amount of mRNA for IFN-γ synthesis in NK-92 cells is not affected by the applied ethanol concentrations as well. Additionally it was shown within the current work, that the reduced IFN-γ production by NK-92 cells in the presence of ethanol might not be explained by an intracellular accumulation of the IFN-γ protein. The inhibitory action of ethanol on IFN-γ may rather be caused by posttranslational modification once IFN-γ is released by NK-92 cells, since the addition of recombinant human IFN-γ to the cell culture supernatants of ethanol-treated cells led to a decline in the amount of IFN-γ concentration. We therefore hypothesized that ethanol may cause the release of either an IFN-γ-binding or IFN-γ-degrading protein. An increase in soluble IFN-γ receptor as a result of ethanol treatment was not observed. But the addition of mixture of 5 commercially available protease inhibitors counteracted the effect of ethanol treatment, giving us a first hint of IFN-γ-modulatory mechanism, where IFN-γ released by NK-92 cells may be disintegrated by a protease released as consequence of ethanol incubation. To our best knowledge we are the first to demonstrate a posttranslational modification of IFN-γ as a consequence of ethanol incubation. In summary, the present results support the inhibitory role of ethanol on IFN-γ, but are too preliminary to explain the underlying immunmodulatory effect.Die Bildung von Zytokinen und damit die Immunabwehr selbst kann durch exogene Faktoren, wie Alkohol beeinträchtigt werden. Der Einfluss von Alkohol auf die Immunabwehr unterscheidet sich je nach Menge des konsumierten Alkohols und des damit erreichten Blutalkoholspiegels, sowie nach der Dauer des chronischen Missbrauchs. In der nun vorliegenden Arbeit wurden die immunmodulatorischen Eigenschaften von Alkohol in Konzentrationen von 1 ? und 3 ? untersucht. Hierzu wurde die Fähigkeit isolierter humaner PBMC unter dem Einfluss von Ethanol pro- bzw. antiinflammtorische Zytokine zu produzieren untersucht. In unserem Modellsystem wurde IFN-γ in seiner Ausschüttung am deutlichsten gehemmt. Hohe Alkoholkonzentrationen von 3 ? führten zu einem hochsignifikanten Rückgang auf ca. 4 % der IFN-γ Menge, wie sie unter alkoholfreien Bedingungen gemessen werden konnte. Für Alkohol-Konzentrationen von 1 ? betrug dieser Rückgang ca. 30 %. Bei der Erstreaktion des Immunsystems wird IFN-γ hauptsächlich von NK Zellen gebildet. Dabei ist IL-12 der effektivste Aktivator der IFN-γ Genexpression und damit entscheidend für eine angemessene Reaktion gegen eingedrungene Pathogene viraler oder bakterieller Genese. Wir konnten belegen, dass die IFN-γ Ausschüttung durch IL-12-stimulierte NK-92 Zellen durch Ethanol in gleicher Weise gehemmt wird, wie bereits für PBMC beschrieben. Diese Hemmung geht jedoch nicht mit einer Veränderung der STAT-4-Phosphorylierung, einem entscheidenden Schritt hin zur IFN-γ-Genexpression, einher. Parallel zu dieser Messung konnte auch keine Verminderung der IFN-γ-mRNA-Menge unter dem Einfluss von Ethanol nachgewiesen werden. Auch eine intrazelluläre Akkumulation kann nach unseren Ergebnissen ausgeschlossen werden. Damit ist die inhibierende Wirkung von Alkohol auf posttranslatorisch wirksame Mechanismen zurückzuführen. Dabei zeigte sich, dass ein unbekanntes Agens im Zellkulturüberstand von zuvor mit Ethanol behandelten Zellen die Fähigkeit besitzt extern zugesetztes humanes rekombinantes IFN-γ so zu modulieren, dass ein anschließender Nachweis im ELISA nur noch in geringen Mengen möglich war. Damit scheint Alkohol die NK-92 Zellen zur Ausschüttung eines IFN-γ bindenden oder IFN-γ abbauenden Moleküls zu stimulieren. Die Bindung an den löslichen IFN-γ-Rezeptor kann nach den Ergebnissen unserer Untersuchungen ausgeschlossen werden. Durch den Einsatz einer kommerziell erhältlichen Mischung von 5 Protease-Inhibitoren konnte der ethanolbedingte Abbau von IFN-γ in den Zellkulturüberständen gehemmt werden und damit ein möglicher Wirkmechanismus aufgezeigt werden

    Posttranslationale Modifikationen wie Glykosylierung oder Palmitoylierung, aber auch Wechselwirkungen mit anderen Proteinen können die Lokalisation von Rezeptoren innerhalb von Detergenz-unlöslichen Membrandomänen/ lipid rafts und somit die Signaltransduktion beeinflussen. In dieser Arbeit wurden die posttranslationalen Modifikationen von LIFR und gp130 und ihr Einfluss auf die Assoziation mit Detergenz-resistenten Membranmikrodomänen untersucht. Palmitoylierung von Cysteinresten innerhalb der Transmembrandomäne eines Proteins kann die DRM-Assoziation beeinflussen. Da gp130 zwei Cysteinreste in der Transmembrandomäne enthält, wurde die Rolle dieser Cysteine für die DRM-Assoziation untersucht. Aus den Befunden kann zusammengefasst werden, dass die Cysteine C711 und C725 in der Transmembrandomäne von gp130 keinen signifikanten Einfluss auf die DRM-Assoziation haben. Nach der Isolierung der DRM mit Brij 58 und Triton X-100 wurde entgegen der Erwartungen sogar eine leichte Steigerung der DRM-Assoziation der C->A-Mutanten beobachtet. Für LIFR konnte nach DRM-Isolierung mit Brij 58 und Triton X-100 eine partielle Assoziation mit den Detergenz-unlöslichen Membrandomänen bestätigt werden. Weiterhin wurden zwei unterschiedliche N-Glykosylierungsformen des Rezeptors nachgewiesen. Die Mannose-reiche (Vorläufer) Form tritt bevorzugt in der Detergenz-löslichen Fraktion auf und wird von dem Enzym Endo-Glykosidase Hf degradiert. Die hybride (reife) Form neigt zur stärkeren Assoziation mit den Detergenz-unlöslichen Mikrodomänen. Nur die reife LIFR-Form wird nach der Bindung von LIF an den Rezeptorkomplex in 3T3-L1- und HepG2-Zellen phosphoryliert, was in Verbindung mit anderen Ergebnissen unserer Arbeitsgruppe darauf hindeutet, dass nur die reife Form an der Zelloberfläche exprimiert wird und an der Signaltransduktion beteiligt ist. In HepG2-Zellen wurde nach Stimulation mit LIF eine Erhöhung der LIFR-Phosphorylierung in Detergenz-unlöslichen Membranmikrodomänen (DRM) nachgewiesen. Phosphoryliertes gp130 wurde nach LIF-Gabe hingegen hauptsächlich in der nicht-DRM-Fraktion detektiert. Die Widersprüchlichkeit dieses Ergebnisses kann auf methodische Probleme zurückgeführt werden. Außerdem wurde in 3T3-L1-Zellen eine stärkere Phosphorylierung des LIFR in der DRM-Fraktion nicht bestätigt. In 3T3-L1 findet die Aktivierung von gp130 und LIFR in der gleichen Plasmamembrandomäne (nicht-DRM) statt. Das deutet einerseits auf zelluläre Unterschiede bezüglich Rezeptoraktivierung und Verteilung innerhalb der Plasmamembran, und andererseits auf unterschiedliche Sensitivität der lipid rafts gegenüber den verwendeten Detergenzien hin.Post-translational modification of proteins is an important event in the regulation of cellular functions. Glycosylation or palmitoylation, but also ligand binding can affect the localization of proteins in membrane microdomains and thus affect signal transduction. The aim of this study was to analyze how posttranslational modifications of LIFR and the common signal transducer gp130 impact the translocation to detergent resistant membranes (DRMs, lipid rafts). Palmitoylation of cysteine residues within the transmembrane domain of a protein is considered to be one process that assists in the localization of proteins to DRMs. Gp130 has two cysteine residues C711 and C725 in its transmembrane domain. My studies indicate that these cysteine residues have no significant influence on lipid raft association of gp130. Contrary to our expectations, after isolation of DRMs with Brij 58 and Triton X-100 an increase of raft association of the C->A-mutants was detected. Partial DRM association of LIFR was confirmed by using Brij 58 and Triton X-100 protocols. Furthermore, two different N-glycosylation types of that receptor could be detected. The mannose-rich (precursor) species is preferentially found in non-DRMs and is degraded by Endo-Glycosidase Hf. The hybrid-type (mature) tends towards an association with DRMs. My results indicate that only the mature-type of LIFR was phosphorylated after LIF binding to the receptor complex in 3T3-L1 and HepG2 cells. Combined with other data from our workgroup these findings suggest that only the mature-type of LIFR is expressed at the plasma membrane surface and involved in signal transduction. After stimulation with LIF an increase of LIFR tyrosine phosphorylation was observed in DRMs in HepG2 cells. However, phosphorylation of gp130 was detected only in non-DRMs fractions after stimulation with LIF. The inconsistency of these results can be explained with methodical problems. Furthermore, the translocation of phosphorylated receptors described above could not confirmed in 3T3-L1 cells. In this cell line, the activation of gp130 and LIFR occurs in detergent-resistant membranes. These findings indicate differences between cell lines with respect to receptor activation and translocation within the plasma membrane on the one hand and demonstrate a differential sensitivity of raft subdomains to extraction by different detergents on the other hand

    Linkages between poverty and sustainable agricultural and rural development in the uplands of Southeast Asia

    Most of the upland areas of Southeast Asia are characterized by insufficient infrastructure, low productivity in smallholder crop and animal production, mounting environmental problems such as soil and forest degradation and loss of biodiversity, increasing population pressure, and widespread poverty, particular in rural areas. While some upland areas in South East Asia have been experiencing considerable progress during the past twenty years, others have stagnated or even declined with respect to economic, social and environmental objectives of development. The purpose of the paper is to describe major trends regarding sustainable development in the upland areas of selected countries in South East Asia, and review explanatory approaches for the observed trends based on case studies from Cambodia, Laos, Thailand, Vietnam, and Indonesia. The conceptual framework for this paper builds on the critical triangle of sustainable rural development. Here, equity or poverty alleviation, economic growth, and the protection of the environment are the three major policy objectives. We further distinguish three explanatory approaches for land use change and agricultural and rural development. Apart from the market approach and the population approach, we suggest that future studies should focus more on governance issues as a major driving force of land use change. The governance approach appears particularly relevant for upland areas which are often politically and institutionally marginalized. The paper begins with a review of definitions of sustainability, and proceeds with a conceptual analysis of the two-way linkages between poverty and the environment, and poverty and economic growth in rural areas. This is followed by empirical findings from research on agriculture and forestry as the major land uses in upland areas of selected South East Asian countries. Based on the results of different case studies from Cambodia, Laos, Vietnam and Indonesia, we seek to contrast stories of relative success with those of failure. The paper concludes with implications for rural and agricultural development policies, and suggests future areas of research

    Studies on the biogenesis of proteins in the mitochondrial inner membrane

    Mitochondrien und Prokaryonten weisen vielerlei Ähnlichkeiten auf, vermutlich sind sie in der Evolution aus gemeinsamen Vorläufern hervor gegangen. So ist zu erwarten, dass sich ausgeprägte Ähnlichkeiten auch in der Biogenese ihrer Proteine nachweisen lassen. In der vorliegenden Studie wurde dieser Vermutung in Untersuchungen zur Biogenese bestimmter Proteinkomplexe der mitochondrialen Innenmembran nachgegangen. Als Modellorganismus diente dabei die Bäckerhefe Saccharomyces cerevisiae. (1) Die γ-Untereinheit der ATP-Synthasen ist in Mitochondrien und Prokaryonten hoch konserviert. Aus älteren Untersuchungen ist bekannt, dass Deletionen am N- oder C-Terminus der γ-Untereinheit nur eine überraschend geringe Reduktion der enzymatischen Aktivität zur Folge haben. In der Experimenten der vorliegenden Studie wurde nun gefunden, dass N- und C-Terminus der γ-Untereinheit für eine effiziente Assemblierung in der ATP-Synthase in den Mitochondrien der Hefe essentiell sind. Bei einer Deletion von 9 Aminosäuren am N-Terminus oder 10 Aminosäuren am C-Terminus wurde die γ-Untereinheit effizient in die Mitochondrien importiert. Der Anteil der Untereinheit, der dann innerhalb von 10 Min. bei 25°C assembliert wurde, reduzierte sich aber um etwa die Hälfte. Deletionen von mehr als 9 N-terminalen oder mehr als 20 C-terminalen Aminosäuren reduzierten den Anteil der assemblierten Untereinheit um mehr als 90%. Hefe-Stämme, die eine verkürzte γ-Untereinheit synthetisierten, waren auf Glycerin als Kohlenstoffquelle nicht lebensfähig. Vermutlich sind N- und C-Terminus der Untereinheit in den ATP-Synthasen sowohl der Mitochondrien, als auch in der Prokaryonten für die Assemblierung von größerer Bedeutung als für die Energieübertragung. (2) Die Metabolittranslokatoren der mitochondrialen Innenmembran sind vermutlich erst im Kontext der Evolution der eukaryontischen Zellen entstanden. Gemeinsam ist den Metabolittranslokatoren das Sequenzmotiv P x (D/E) x x (K/R), das als Carrier signature bezeichnet wird. Für ein Mitglied der Proteinfamilie, den Dicarboxylattranslokator, wurde nun gefunden, dass die Carrier signature die Biogenese des Proteins wesentlich erleichtert. Insbesondere wird der Transport des neu synthetisierten Proteins vom Cytosol in den Intermembranraum wesentlich beschleunigt. (3) Das Protein Oxa1 gehört zu einer Proteinfamilie, zu der auch das Protein YidC der Prokaryonten zählt. Sowohl Oxa1, als auch YidC sind Mediatoren der Proteininsertion in ihren jeweiligen Membranen, und in dieser Funktion u.a. an ihrer eigenen Biogenese beteiligt. Eine Reihe verschiedener Experimente zur Biogenese des mitochondrialen Oxa1 hat nun ergeben, dass neu synthetisiertes Oxa1 nach Import in die Mitochondrien wahrscheinlich nicht vollständig in die mitochondriale Matrix transportiert wird. Vielmehr scheint es im TIM23-Komplex, der Proteintranslokase der Innenmembran, zu akkumulieren und dann unmittelbar in die Lipidphase der Membran zu inserieren. Damit zeigt das Oxa1 eine ähnliche Biogenese wie das bakterielle YidC, das zunächst von der SecYEG-Translokase aufgenommen und dann unmittelbar in die Plasmamembran eingelagert wird. Oxa1 und YidC scheinen somit nicht nur in ihrer Struktur, sondern auch in ihrer Biogenese und in ihrer Funktion signifikante Ähnlichkeiten zu haben. Insgesamt zeigte sich in den Experimenten der vorliegenden Studie, dass sich mitochondriale und prokaryontische Proteine auch nach mehr als zwei Milliarden Jahren getrennter Evolution selbst in molekularen Details ihrer Funktion noch sehr ähnlich geblieben sind.Mitochondria and prokaryotes show many similarities and it is a well established notion that they have common ancestors. It is therefore reasonable to expect significant similarities also in the biogenesis of their proteins. This study followed this idea in investigations on the biogenesis of protein complexes in the mitochondrial inner membrane. The yeast Saccharomyces cerevisiae served as a model organism. (1) The γ-subunit of ATP synthases is highly conserved both in mitochondria and in prokaryotes. Previous studies demonstrated that deletions at the N- or C-terminus of the subunit entail only mild reductions in the enzymatic activity, and the reason of the conserved structure was enigmatic. The experiments of this study show that N- and C-terminus of the γ-subunit are essential for an efficient assembly in the ATP synthase. A deletion of 9 residues at the N-terminus or 10 residues at the C-terminus reduced the ratio of the subunit that assembled within 10 min. at 25°C by about 50%. Deletions of more than 9 N- or more than 20 C-terminal residues reduced the share of the assembled subunit by more than 90%. Yeast strains that synthesized a shortened γ-subunit did not grow on glycerole. N- and C-terminus are probably more relevant for assembly of the ATP synthase than for the transmission of energy. It is proposed that this is the case both for mitochondria and for prokaryotes. (2) The metabolite carrier proteins of the mitochondrial inner membrane were probably newly developed during the evolution of the eukaryotic cells. A common sequence motif of all carrier proteins is the carrier signature, P x (D/E) x x (K/R). The data of this study show that the carrier signature substantially facilitates the biogenesis of the dicarboxylate carrier (DIC). In particular, the translocation of this protein across the mitochondrial outer membrane is significantly accelerated. (3) The mitochondrial inner membrane protein Oxa1 is a member of a protein family that includes the bacterial protein YidC. Both Oxa1 and YidC act as mediators of protein insertion in their membranes, and both proteins participate in their own biogenesis. In this study, a series of experiments indicates that newly synthesized Oxa1 is not imported into the mitochondrial matrix, but accumulates in the inner membrane TIM23 translocase, for direct integration into the lipid bilayer. Oxa1 thereby shows a similar principle of membrane insertion as prokaryotic YidC. This was previously shown to first accumulate in the SecYEG translocase and then to directly integrate into the bacterial plasmamembrane. Oxa1 and YidC thus seem to resemble each other both in their structure and in their biogenesis.
